Two known mutagenic drugs, cyclophosphamide and vinblastine sulfate, were tested using the micronucleus test in the native fish species, Astyanax bimaculatus, in order to determine which of these drugs and the doses which would be the most adequate for use as positive controls in this species. This Brazilian fish species was chosen because few toxicity studies have used native fish species and this particular species is widely consumed in various regions of Brazil. Three thousand erythrocytes per specimen were scored. Doses of 16 and 8 mg/kg body weight of cyclophosphamide and vinblastine sulfate, respectively, were the most effective in causing micronuclei. Cyclophosphamide was the most mutagenic of the two drugs and is recommended for use as a positive control in A. bimaculatus.
Duas drogas reconhecidas como mutagênicas, ciclofosfamida e vimblastina sulfato, foram avaliadas usando o teste do micronúcleo em uma espécie de peixe nativa, Astyanax bimaculatus, para detectar que droga e quais doses são as mais adequadas para serem usadas como controles positivos para esta espécie. Esta espécie de peixe brasileira foi escolhida devido à escassez de estudos toxicológicos com espécies de peixes nativos e também porque ela é amplamente consumida em algumas regiões do Brasil. Um total de 3000 eritrócitos por espécimen foram contados. As doses de 16 e 8 mg/kg de peso corporal de ciclofosfamida e de vimblastina sulfato, respectivamente, foram as mais efetivas na indução de micronúcleos. A ciclofosfamida mostrou ser o melhor agente mutagênico para ser usado como um controle positivo para Astyanax bimaculatus

In the present work the induction of micronuclei in erythrocytes of Cyprinus carpio treated with X-rays and colchicine is studied for the evaluation of mutagenic effects of both clastogenic and mitoclastic (spindle poisoning) agents in this system.Three different experiments were performed treating groups of laboratory-reproduced animals with (1) single doses of X-rays (0.1, 0.5 and 2 Gy); (2) a single i.p. injection of colchicine at the concentrations: 1.6 × 10 −2 , 8× 10 −2 , 0.4 and 2 mg/kg b.w. so as to mimic an acute exposure to the agent and (3) six repeated i.p. injections of the first three concentrations of colchicine, over a period of 18 days, so as to mimic a chronic exposure. Repeated blood samplings were performed by cardiac puncture over a period of about 2 months after the treatment and micronucleus frequencies were determined at multiple times on the same individuals after mutagen exposure. A dose-dependent increase in the micronucleus frequency was observed in irradiated fish and a peak value detected at 21 days. Slight increases of micronucleus frequencies were also observed in both colchicine experiments only for the highest concentrations at the earliest sampling time. Higher concentrations of colchicine clearly showed a lethal effect. According to the present data the micronucleus frequency induced by the highest colchicine dose is comparable to that observed after 0.1 Gy of X-ray irradiation.

Fish have been shown to act as sentinel organisms in genetic toxicology studies and the micronucleus (MN) test using fish erythrocytes has been successfully used to evaluate the genotoxic impact of environmental pollutants on aquatic environments. This study used the MN test of peripheral blood of specimens of a native fish species (Hyphessobrycon luetkenii) to evaluate the genotoxicity of the water of two unconnected reservoirs of the Canela National Forest in southern Brazil. Fish were caught in the reservoirs and MN frequencies in blood smears were determined. Baseline MN frequency of H. luetkenii was determined in the uncontaminated reservoir (reference site) and compared with the results obtained at two different sampling periods (winter and spring) in the reservoir which receives discharges of industrial and municipal wastes. In the spring season, MN frequency in the contaminated reservoir was about five times higher than in the reference site (0.51% and 0.10%, respectively). The results of this study indicate that some fish populations are at risk of exposure to genotoxic pollutants in the Canela National Forest.
