Fire blight resistance of apple, pear and quince genetic resources from Lake Van Basin (eastern Turkey) was tested using Erwinia amylovora strain Ea Van. Shoot tips of 92 native accessions (48 accessions for apple, 38 accessions for pear and 6 accessions for quince) were wounded for inoculation, and artificially inoculated with pathogenic bacteria under greenhouse conditions. The levels of resistance of accessions were classified in comparison with control varieties according to the genotype susceptibility index (GSI%) scores based on the lesion length on shoots of each genotype. Fire blight resistance of accessions consisted of five classes: resistant (R), moderately resistant (MR), moderately susceptible (MS), susceptible (S) and highly susceptible (HS). GSI% scores differed significantly among accessions from each fruit species (p<0.01). GSI values ranged from 12.4% to 64.1% for apple genotypes, from 17.2% to 55.1% for pear genotypes, and from 17.8% to 43.4% for quince genotypes. No resistant genotypes of apple, pear and quince were observed. Seven accessions of apple, two accessions of pear and one accession of quince were MR. 25 accessions of apple, 14 accessions of pear and one accession of quince were MS. These findings indicate a considerable variation in fire blight resistance and could contribute to breeding efforts regarding fire blight resistance in apple, pear and quince.

Sixty-four apple cultivars and selections of potential interest to apple producers and plant breeders in the Czech Republic were tested for their relative resistance to the fire blight pathogen over six years. Level of fire blight resistance was evaluated according to the extent of lesion development on the shoots tips after artificial inoculation in experimental plots under insectproof nets. Cultivars Quinte (resistant) and Yellow transparent (high susceptible) were included in the tests. Of 64 apple cultivars and selections tested, none were high resistant, 3.1% were evaluated as resistant, 10.9% moderately resistant, 57.8% moderately susceptible, 21.9% susceptible and 6.3% high susceptible. Resistant apple genotypes, showing blight necrosis of shoots of 11–12%, were only cultivars Selena and Quinte. Moderately resistant genotypes (blight necrosis 13.1–25.0%) were Kordona, Golden Smoothee, Julia, HL 323, Melodie, HL 421 and S 634/3. High susceptible genotypes (blight necrosis more then 80.1%) were comprised cultivars Vesna, Topas, Yellow transparent and Vanda. The remaining genotypes were moderately susceptible (blight necrosis 26.1–60.0%) and susceptible (blight necrosis 60.1–80.0%). During six experimental years, quantitative variability was recorded in the blight score. Differences between cultivars in susceptibility to fire blight were often statistically significant.

Phenotypic and marker-assisted characterization of new apple genotypes with high resistance to fire blight

In the period 2016–2018 the susceptibility to fire blight of seven apple genotypes, including four recently bred was assessed. The studies showed that two of the genotypes (J-2003-05 and ‘Early Szampion’) are highly resistant to the disease. The indicator of assessment was evaluated by the size of necrotized part of terminal shoots after their inoculation with strain Ea659 of Erwinia amylovora. The analysis of disease incidence on individual shoots of both genotypes showed no symptoms on 92% and 100% of shoots inoculated in 2016, 83.8 and 77.0% of shoots in 2017, and 87.8 and 77.3% of shoots inoculated in 2018, respectively. Molecular tests based on 16 genetic markers revealed that genotype J-2003-05 possesses 10 resistance alleles (covering the region of interest from LG 3, 5, 7, 10 and 12). This genotype is derived from the cross between ‘Melfree’ and ‘Sawa’ and produces very large dark-red tasty fruits. Genotype ‘Early Szampion’ representing the selected hybrid from the cross of ‘Gold Milenium’ and ‘Šampion’, producing large, bright red, attractive fruits, was found to possess 10 resistance alleles (covering the region of interest from LG 3, 7, 10 and 12), Both genotypes have the prospect of being used in breeding programs and commercial planting.
