Despite the importance of field-grown cucurbits in Spain, only limited information is available about the impact of disease on their production. During the 2003 and 2004 growing seasons, systematic surveys were carried out in open field melon (Cucumis melo) and squash (Cucurbita pepo) crops of Murcia Province (Spain). The fields were chosen with no previous information regarding their sanitation status, and samples were taken from plants showing viruslike symptoms. Samples were analyzed using molecular hybridization to detect Beet pseudo-yellows virus (BPYV), Cucurbit aphid-borne yellows virus (CABYV), Cucumber mosaic virus (CMV), Cucumber vein yellowing virus (CVYV), Cucurbit yellow stunting disorder virus (CYSDV), Melon necrotic spot virus (MNSV), Papaya ringspot virus (PRSV), Watermelon mosaic virus (WMV), and Zucchini yellow mosaic virus (ZYMV). We collected 924 samples from 48 field plots. Out of these, almost 90% were infected by at least one of the viruses considered, usually CABYV, which was present in 83 and 66% of the melon and squash samples, respectively. In the case of melon, CYSDV, BPYV, and WMV followed CABYV in relative importance, with frequencies of around 20 to 30%, while in squash, CVYV and BPYY showed frequencies between 28 and 21%. The number of multiple infections was very high, 66 and 56% of the infected samples of melon and squash, respectively, being afflicted. CABYV was present in all multiple infections. The high incidence of CABYV in single and multiple infections suggests that this virus may well become an important threat for cucurbit crops in the region. Restriction fragment length polymorphism (RFLP) analysis revealed that CABYV isolates can be grouped into two genetic types, both of which seemed to be present during the 2003 epidemic episode, but only one of the types was found in 2004.

In Venezuela, cucurbit viruses have been associated with important yield losses. Therefore, an extensive survey was conducted to determine the major cucurbit viruses in this country. Leaf samples from 284 cucurbit plants exhibiting virus-like symptoms were collected mainly in 2009-2010 from several states of Venezuela. They were assessed for viral infection by polymerase chain reaction (PCR) for Melon chlorotic mosaic virus (MeCMV) and reverse transcriptase (RT)-PCR for Papaya ringspot virus (PRSV), Zucchini yellow mosaic virus (ZYMV), Watermelon mosaic virus (WMV), Cucumber mosaic virus (CMV), Squash mosaic virus (SqMV) and Cucurbit aphid-borne yellows virus (CAB-YV). The most common virus in cucurbit fields, MeCMV, was present in 65Á8% of samples. Its associated alphasatellite was found in 78% of samples positive for MeCMV. PRSV, ZYMV and WMV were found with different prevalence: 34Á2, 32Á4 and 1Á1% respectively. CMV was also detected (6Á7%) but SqMV and CABYV were not found. Single infections were more frequent than mixed infections (56Á4 and 38Á6%, respectively). For ZYMV, comparison and phylogenetic analyses of either polymerase and coat protein (NIb-CP) partial sequences or CP complete sequences revealed a low genetic diversity within Venezuelan isolates. Thirty-four ZYMV isolates were used for serological and biological analysis. Thirteen monoclonal antibodies showed a major group of isolates spread in several states and two groups located in Zulia only. Venezuelan ZYMV isolates showed biological variability on cucurbit cultivars susceptible, tolerant or resistant to ZYMV. Resistance to ZYMV in cucumber appears potentially durable, whereas resistance or tolerance in zucchini and melon may be easily overcome.

During 2010, yellowing symptoms were frequently observed in cultivated squash fields in Egypt. A total of 717 symptomatic squash leaf samples were collected from four regions where squash cultivation is of economic importance for the country: Kafrelsheikh, El-Behira, El-Sharkia and El-Ismailia. Serological analysis showed that 95.6% of the symptomatic squash samples were infected by Cucurbit aphid-borne yellows virus (CABYV), and visual estimation of the incidence of yellowing symptoms suggested a very high incidence of CABYV in the fields. Twelve CABYV isolates were characterized by sequencing two regions of the viral genome, open reading frame (ORF) 3 and ORFs 4/5. Overall, Egyptian isolates were very similar among them, and had higher similarity values with a French than with a Chinese isolate. The average nucleotide diversity for ORF 3 was significantly higher than for the other two regions, indicating that variability is not evenly distributed along the viral genome. The ratios between nucleotide diversity values in non-synonymous (d N ) and synonymous (d S ) positions (d N /d S ) for each ORF showed that the three ORFs are evolving under different pressures, although predominantly under purifying selection. Phylogenetic analyses revealed that these Egyptian isolates, with only one exception, shared the same clade with a French isolate. Moreover, these analyses suggested that Egyptian isolates belong to the Mediterranean group described previously.
