Anciently introduced species can be confounded with native species because introduction pre-dates the first species inventories or because of the loss of the collective memory of the introductions. The term 'cryptogenic species' denotes species of unknown or unclear status (native versus non-native) in a given territory, and disciplinary approaches are often insufficient for solving their true status. Here, we follow an integrative, multidisciplinary approach to solve the status of a cryptogenic species, proposing that building on evidence from multiple disciplines can produce robust and clarifying insights. We undertook an exhaustive review of information on a putatively native crayfish (Austropotamobius italicus) in Spain. The reviewed information included taxonomy, genetics and phylogeography, history, archaeology, linguistics, biogeography, ecology, symbiotic organisms and even gastronomy and pharmacy. The knowledge produced by different scientific disciplines converges to indicate that A. italicus is a non-native species in Spain. Historical documents even identify the first introduction event: crayfish were shipped from Italy to Spain in 1588 as a diplomatic gift from Francesco I de' Medici to King Philip II of Spain. Previous discussions on the status of A. italicus focussed on inconclusive and often confusing genetic results and excluded the rich and clarifying evidence available from other approaches and disciplines. Interdisciplinarity is an often-invoked but rarely implemented practice in an academic environment that increasingly promotes narrowfocussed specialization. Our review shows that the integration of disciplines can surpass disciplinary approaches in solving scientific controversies. Our results have straightforward implications for strategies to conserve biological diversity in Spain and Europe, urging a debate on the appropriateness of devoting conservation efforts to nonnative species.

Crayfish of North American origin are amongst the most prominent high-impact invasive invertebrates in European freshwaters. They contribute to the decline of European native crayfish species by spreading the pathogen causing crayfish plague, the oomycete Aphanomyces astaci. In this study we validated the specificity of four quantitative PCR (qPCR) assays, either published or newly developed, usable for environmental DNA (eDNA) screening for widely distributed native and non-native crayfish present in Central Europe: Astacus astacus, Pacifastacus leniusculus, Faxonius limosus and Procambarus virginalis. We then conducted an eDNA monitoring survey of these crayfish as well as the crayfish plague pathogen in a wide variety of habitat types representative for Central and Western Europe. The specificity of qPCR assays was validated against an extensive collection of crayfish DNA isolates, containing most crayfish species documented from European waters. The three assays developed in this study were sufficiently species-specific, but the published assay for F. limosus displayed a weak cross-reaction with multiple other crayfish species of the family Cambaridae. In the field study, we infrequently detected eDNA of A. astaci together with the three non-native crayfish species under examination. We never detected eDNA from A. astaci together with native crayfish, but in a few locations eDNA from both native and non-native crayfish was captured, due either to passive transport of eDNA from upstream populations or co-existence in the absence of infected crayfish carriers of A. astaci. In the study, we evaluated a robust, easy-to-use and low-cost version of the eDNA sampling equipment, based mostly on items readily available in garden stores and hobby markets, for filtering relatively large (~5 l) water samples. It performed just as well as the far more expensive equipment industrially designed for eDNA water sampling, thus opening the possibility of collecting suitable eDNA samples to a wide range of stakeholders. Overall, our study confirms that eDNA-based screening for crayfish and their associated pathogen is a feasible alternative to traditional monitoring.

-The white-clawed crayfish is a complex of species (Austropotamobius pallipes plus A. italicus, Crustacea) pivotal to riverine environments. Regrettably, it is endangered in many European countries due to a recent massive decline. We carried out a fine-scale survey on white-clawed crayfish distribution, morphology and genetics in an under-investigated region of central Italy (Lunigiana, Tuscany) to improve local knowledge and aid conservation management of the species. The torrential fauna of Lunigiana is scarcely known and habitat alterations due to the anthropic exploitation of the territory represent a potential threat to its persistence. We investigated crayfish occurrence in eight streams of the Magra River Basin. We performed nocturnal and diurnal transects, took morphometric measurements, collected samples for genetic analysis and checked for the occurrence of ectosymbionts. Crayfish were disclosed in two streams (Civasola, Verdesina). Morphometric and phylogenetic analysis (mtDNA COI gene Bayesian tree reconstruction) concurrently identified both populations as Austropotamobius italicus carinthiacus. In the Verdesina stream, crayfish were heavily infested by branchiobdellidans (Annelida). Verdesina crayfish were also significantly smaller than those found in the Civasola stream, where branchiobdellidans were absent. Hence, we hypothesized that such difference in size might be related to the high density of ectosymbionts. In the light of habitat features and of data herein provided, we propose that both Civasola and Verdesina streams should be considered as "natural ark sites" for A. italicus, with the Magra River Basin representing a regional stronghold for the conservation of the species.Keywords: astacidae / branchiobdellida / cytochrome oxidase I / DNA barcoding / Austropotamobius fulcisianus Résumé -Données préliminaires sur la distribution, la morphologie et la génétique des écrevisses à pattes blanches et leurs ectosymbiotes en Lunigiana (Toscane, Italie).
