Following a request from the European Commission, the Panel on Plant Health performed a pest categorisation on tomato leaf curl New Delhi virus (ToLCNDV). ToLCNDV is a well‐defined bipartite Begomovirus species, sometimes associated with satellite molecules. It is transmitted by Bemisia tabaci to a wide range of hosts. ToLCNDV is reported from Estonia, Greece, Italy, Portugal and Spain, with limited distribution. The prevalent strain (ToLCNDV‐ES) in these countries is particularly adapted to cucurbits and is different from isolates reported outside the EU, which are better adapted to solanaceous crops and could therefore pose additional risk for EU agriculture. The virus is regulated under Commission Implementing Regulation (EU) 2019/2072. The main pathway of entry identified is plants for planting of susceptible hosts, even if entry could also occur via commodities carrying viruliferous B. tabaci and possibly by seeds. While establishment and local spread rely on B. tabaci, the virus can also be dispersed over long distances by movement of infected plants for planting. Establishment and spread are limited to regions with ecoclimatic conditions suitable for the establishment of vector populations (southern regions of Europe) or can occur as outbreaks wherever crops are grown under protected cultivation. The main uncertainties associated with this pest categorisation are the distribution and prevalence of ToLCNDV in the EU, the magnitude of the virus impact particularly on hosts different from Cucurbitaceae, and seed transmission. ToLCNDV meets all the criteria evaluated by EFSA to qualify as potential Union Quarantine Pest (QP); conversely, ToLCNDV does not meet the criterion of being widespread in the EU to qualify as a Regulated Non‐Quarantine Pest (RNQP). Should new data show that ToLCNDV is widespread in the EU, the possibility would exist for non‐EU isolates to qualify as QP, while ToLCNDV EU isolates (ToLCNDV‐ES) could qualify as RNQP.

Tomato leaf curl disease is a serious constraint to tomato cultivation across the globe. The disease is characterized by leaf curling, crinkling, stunting and puckering with the drastic reduction in plant growth and yield. During two‐year survey in major tomato‐growing belt of Tarai region located in foothills of Himalayas in Uttarakhand, India, the mean incidence and per cent disease index of the disease were found to be 68.76% and 67.19%, respectively. In the morphological analysis using electron microscopy, pathogen contains twin geminate particles measuring 30 × 20 nm. The PCR amplification using RUGEMF1 and RUGEMR1 primers and sequencing of the amplicons, revealed more than 97% similarity to Tomato leaf curl New Delhi Virus (ToLCNDV). Thus, based on the sequence similarity, morphology and whitefly transmission, begomovirus isolate was recognized as a Tomato leaf curl New Delhi virus and designated as ToLCNDV_[Pant]. Amplification and sequencing of the Bemisia tabaci partial mtCOI gene from the Tarai region revealed the prevalence of Asia‐II‐1 cryptic species in the region. The overall mean pairwise distance across the 10 sequences studied was 0.0139, whereas the reference Asia‐II‐1 distance ranged from 0.0031 to 0.0332. The current investigation will aid in the development of an integrated strategy to ToLCD management.

Tomato leaf curl New Delhi virus (ToLCNDV) is a whitefly-transmitted bipartite ssDNA virus causing worldwide havoc to the tomato production. The present study addresses first report for the existence of ToLCNDV at Manipur region of North East India. Sequence comparison of DNA A (MG649330) and DNA B (OM752176) genomes showed close homogeneity of about 98% and 100% with Bangladesh isolates, but revealed only 84–96% (DNA A) and 81–100% (DNA B) similarity with other isolates of ToLCNDV. Phylogenetic relationship of global ToLCV species showed interchangeable results, substantiating that the isolate belonged to the old world bipartite group. Significant recombination events were ascertained around the intergenic region and the 5’end of the AC1 gene implicating a constant driving force for evolution and emergence of the strain. Population dynamics of global ToLCNDV isolates and other dominant ToLCV species within India were determined based upon several parameters i.e. genetic diversity, historic demographic events including neutrality test, Fst distance, Mismatch distribution plot, Haplotype network yielding significant results. Population genetics analysis overall showed low nucleotide diversity indicating recent population expansion. The dominant species of ToLCV in India also reinforced similar results where all the population showed diversifying selection constraint for the coat protein genes. Population dynamics of these viruses portrays Indian subcontinent as the possible hotspot for rapid demographic expansion from a small virus population size, indicating probability for rapid spread and emergence of distinct strains which may assist in future to signal the emergence of new threats to tomato production.
