First Complete Genome Sequence of Currently Circulating Infectious Bronchitis Virus Strain DMV/1639 of the GI-17 Lineage

Abstract: Avian infectious bronchitis virus is the causative agent of a highly contagious disease that results in severe economic losses to the poultry industry worldwide. Here, we report the first coding-complete genome sequence of strain DMV/1639 of the GI-17 lineage, isolated from broiler chickens in Georgia in 2019.

“…Other molecular techniques, such as loop-mediated isothermal amplification (LAMP) and padlock probes (PLPs) combined with rolling circle amplification (RCA), have been validated for IBV detection [ 78 , 79 ]. Additionally, NGS techniques have recently been used to study IBV [ 80 , 81 ]. This approach, despite being currently too expensive and laborious for routine use, has proven useful for research purposes, particularly because it allows to precisely study the presence of subpopulations within a single sample and contextualize IBV within the respiratory disease complex.…”

“…Next-generation paired-end (2 × 150-bp) sequencing was performed on an Illumina MiSeq instrument using the 300-cycle MiSeq reagent kit version 2. The sequence data were assembled using a de novo approach and utilizing MIRA3 version 0.0.1 ( 19 ) within a customized workflow on the Galaxy platform ( 20 ), as described previously ( 21 , 22 ). The MiSeq run generated from 1,303,453 to 2,446,408 total paired-end reads per sample ( Table 1 ).…”

“…Next-generation paired-end sequencing (2 ϫ 250 bp) was performed on an Illumina MiSeq instrument using the 500-cycle MiSeq reagent kit v2 (Illumina, USA). Sequence data were assembled using a de novo approach and utilizing MIRA3 version 0.0.1 (10) within a customized workflow on the Galaxy platform (11), as described previously (12,13). The final outputs of the analysis workflow included a set of summary statistics on the run and the assembly.…”
