First Complete Genome Sequence of Clostridium sporogenes DSM 795 T , a Nontoxigenic Surrogate for Clostridium botulinum, Determined Using PacBio Single-Molecule Real-Time Technology

Abstract: The first complete genome sequence of Clostridium sporogenes DSM 795T, a nontoxigenic surrogate for Clostridium botulinum, was determined in a single contig using the PacBio single-molecule real-time technology. The genome (4,142,990 bp; G+C content, 27.98%) included 86 sets of >1,000-bp identical sequence pairs and 380 tandem repeats.

“…C. botulinum Group I is one of four distinct groups within the species C. botulinum , a polyphyletic taxon united only by the ability to form the highly potent botulinum neurotoxin [ 1 , 2 , 3 , 4 , 5 ]. C. sporogenes has often been perceived as a non-toxic close relative of C. botulinum Group I [ 6 , 7 , 8 , 9 , 10 , 11 , 12 , 13 ]. It is an important cause of food spoilage [ 14 , 15 , 16 ], and used as a surrogate for C. botulinum Group I in food sterilisation tests [ 7 , 14 , 15 , 17 , 18 , 19 , 20 ].…”

“…Preliminary attempts to purify the nitroalkene reductase directly from C. sporogenes by conventional chromatography indicated the presence of more than one enzyme reducing (E) -2-nitro-1-phenylpropene (unpublished results). Furthermore, the analysis of genome sequences of C. botulinum ATCC 3502, C. sporogenes ATCC 15579 and recently published genomes of C. sporogenes NCIMB 10696 [ 39 ] and C. sporogenes DSM 795 [ 40 ] revealed the presence of an open reading frame showing features characteristic for OYE-like reductases ( Fig. 1 ; CLOSPO_03444).…”

“…We have sequenced a large number of genomes using PacBio RS II and published many scientific publications with collaborators in Okinawa genome projects in a wide variety of area including medical area. For viral and prokaryotic genomes, we determined all eight RNA segments of influenza A virus as full-length sequences (data not published), 21 complete bacterial genomic sequences, and 2 entire bacterial biosynthetic gene clusters [ 17 21 , 30 34 , 36 , 41 44 ]. For eukaryotic genomes, we constructed almost-complete (totally 95% of 540 Mb in 11 chromosomes) genome sequences of azuki bean [ 37 ].…”
