Abstract. Melanoma is a devastating disease frequently encountered within both veterinary and human medicine. Molecular changes linked with neoplastic transformation of melanocytes include mutations in genes that encode proteins intrinsic to the regulatory pathways of two tumor suppressor proteins (retinoblastoma protein and p53), proto-oncogene mutation to oncogenes, altered expression of epithelial cadherin and CD44 adhesion molecules, and upregulation of angiogenic factors and other growth factors. Histologic evaluation of the primary mass is the most common means of diagnosis, with cytology used more frequently to document metastasis. Melanoma's highly variable histologic and cytologic patterns can make diagnosis by either method problematic. Adherent epithelioid morphology, including signet ring forms, and nonadherent round and spindle forms are recognized, with pigmentation an inconsistent finding. The site of the tumor, the thickness of the primary tumor or depth of invasion, and the number of mitotic figures per high-power field or per millimeter are used histologically to predict biologic behavior, whereas site and degree of pleomorphism are typically used for cytologic preparations. Diagnosis of amelanotic melanoma can be aided by ancillary diagnostic techniques. Tumor cells are usually positive for vimentin, S100, neuron-specific enolase, and Melan-A, and negative for cytokeratin. Melan-A as a positive marker is not as sensitive as the others are but is likely more specific. Monoclonal antibodies to human melanosome-specific antigens 1 and 5 cross-react with canine antigens for a combined sensitivity rate of 83%. Mouse monoclonal antibody IBF9 specifically recognizes canine melanoma antigen and also has good sensitivity. Serologic markers, including cytokines, cell adhesion molecules, and melanoma-inhibitory activity, are being investigated as potential sentinels of melanoma. Currently, there is no single diagnostic technique capable of differentiating benign from malignant melanocytic neoplasms or predicting survival time.

Background: Cutaneous and subcutaneous metastatic deposits are rare and can be seen in 0.8-5% cases. This study was done to evaluate the role of Fine Needle Aspiration Cytology(FNAC) in the diagnosis of cutaneous and subcutaneous nodules in patients with known malignancy or as a primary manifestation of an unknown malignancy. Methods:It was a 5 year retrospective study and the record of patients subjected to FNAC during this period was reviewed.Results: Out of total number of 13572 patients 58(0.42%) were diagnosed as cutaneous /subcutaneous metastatic deposits .In 51 cases ,the primary was known and in 7 cases it was an initial manifestation of an unknown primary. There were 39 males and 19 females with age range of 27-82 years .Conclusion: Commonest site of metastases was chest wall and most common primary was lung. Adenocarcinoma was the commonest diagnosis on morphology. This study emphasizes the efficacy of FNAC on the accurate diagnosis of subcutaneous nodules especially in patients with known malignancy .It also offers clue to the underlying malignancy in occult primary.

RESUMOObjetivo: Estabelecer um modelo experimental de cultura primária de melanoma metastático por punção aspirativa por agulha fina (PAAF). Métodos: Duas culturas primárias de células de melanoma metastático através de PAAF foram desenvolvidas a partir de pacientes submetidos à excisão de lesões metastáticas e identificadas por imunohistoquímica utilizando os marcadores S-100 e MB-45, específicos para melanoma. Resultados: O diagnóstico da cultura foi confirmado através da imunohistoquímica. A técnica oferece vantagens como pequena morbidade e invasibilidade, além do mais, a mesma lesão pode ser puncionada seqüencialmente durante o tratamento do doente. Conclusão: O estabelecimento de cultura primária de melanoma metastático a partir de punção aspirativa por agulha fina mostrou-se uma técnica viável. Descritores: Melanoma, Excisão de Linfonodos. Results: The culture diagnostic was confirmed by immunohistochemical. The technique of FNAB offers the advantage of providing a sequential analysis of the same tumor nodules throughout treatment, and is a minimally invasive procedure with almost no associated morbidity.
