Filtered platelet concentrates from pooled buffy coats show comparable storagelesions when stored for 9 d at 20–24°C or when supplemented with ThromboSolat 2–6°C

Abstract: The present study investigated the quality of platelet concentrates from pooled buffy coat (PCBC) along different production steps and during storage to characterize storage lesions reflected by platelet activation, changing metabolic and cell turnover status (pH, LDH activity). These criteria were compared in conventionally stored PCBCs (20-24 degrees C, n = 8, Group I) to cold stored PCBCs (2-6 degrees C, n = 8, Group II) when supplemented with ThromboSol. Platelet activation was measured on days 1, 3, 7 and… Show more

“…Of the 19 (58%) articles in which the CSP products were WBD, 10 (53%) articles were produced by buffy coat preparations, and nine (47%) were produced via platelet‐rich plasma (PRP) preparations . The predominant anticoagulant for WBD platelets was citrate–phosphate‐dextrose, which was reported in 10 (53%) articles (Table ) .…”

“…The time interval between platelet donation and the start of cold storage was specified in 20 (60%) of the short‐listed articles . The time interval was reported as being within 6 hours of donation in 10 articles, between 6 and 24 hours in three articles, and more than 24 hours in seven articles (Fig.…”

“…Even under optimal conditions, stored PCs demonstrate a storage lesion characterized by a variety of abnormalities, among them increased expression of surface glycoproteins, 5 whereby only more sensitive methods can document the formerly assumed stability of PLT surface antigens during storage. It was shown earlier that expression of the PLT surface glycoproteins CD41a and P‐selectin are increasing in pooled PCs 6 . It is hypothesized that the amount of ABH antigen expressed on a PLT's surface might be related to its activation status.…”

“…Expression of activated forms of GPIIb‐IIIa have been evaluated in different PCs using a specific antibody (PAC‐1). Results of several studies indicate that PAC‐1–positive platelets are significantly increased in different platelet products, indicating that they have been preactivated . Reduced platelet aggregation should be expected for platelet products showing an enhanced binding of PAC‐1.…”
