Fighting Food Allergy by Inducing Oral Tolerance: Facts and Fiction

Abstract: The prevalence of food allergy (FA) is increasing, and there is an urgent need to take effective measures against it. One important measure is the avoidance diet, which shows a disadvantage, especially in case of accidental exposure. Oral tolerance restoration sheds new light on the control of FA. Oral tolerance is naturally a state of systemic unresponsiveness of the gastrointestinal tract to food antigens and its restoration can be a clinical therapy for FA. Its immune basis lies on the intestinal mucosal im… Show more

“…A mother's distress while breastfeeding can alter milk microbiota ( 33 ) or lower milk secretory immunoglobulin A ( 34 ), both of which affect infant gut immunity ( 35 ). Hence, impaired mother-infant feeding or social interactions may lower socio-emotional responses in the infant ( 36 ) and, via the gut-brain axis, lead to food intolerance ( 37 ). Since parent-child interactions, namely strategies for infant soothing, were not assessed in our study, we are unable to offer explanations for the unexpected findings on food sensitization and improved cognition or language in boys.…”

“…Allergic enterocolitis with food allergies is characterized by severe pain in the abdomen, flatulence, and loose stools with the discharge of vitreous mucus, which contains many eosinophils. Patients with allergic enterocolitis complain of extreme weakness, loss of appetite, headache, and dizziness (9,12,13,73). Histological examination of patients with allergic enterocolitis reveals hemorrhagic changes, pronounced tissue eosinophilia, local edema, and mucus hypersecretion (7,9,68,72,81,84,85).…”

“…The default, "healthy" reaction of the immune system to food proteins is called oral tolerance, which is the continuous unresponsiveness of the immune system to the 130-190 g of food proteins daily absorbed, a process that is largely mediated by the gut-associated lymphoid tissue (GALT) (Brandtzaeg, 2009;Yang et al, 2021). GALT is the largest lymphoid tissue in the body, comprising among others the Peyer's patches, the lymphoid follicles, and the mesenteric lymph node (MLN).…”
