Fidelity in planar cell polarity signalling

Abstract: The polarity of Drosophila wing hairs displays remarkable fidelity. Each of the approximately 30,000 wing epithelial cells constructs an actin-rich prehair that protrudes from its distal vertex and points distally. The distal location and orientation of the hairs is virtually error free, thus forming a nearly perfect parallel array. This process is controlled by the planar cell polarity signalling pathway. Here we show that interaction between two tiers of the planar cell polarity signalling mechanism results … Show more

“…6). This is consistent with the fact that Drosophila mutant phenotypes of both ft and ds have short and thick adult legs, despite the fact that ft and ds have distinct expression patterns in Drosophila imaginal discs (Garoia et al, 2000;Ma et al, 2003). These results were not expected if Ds and Ft acted as a traditional ligand and receptor.…”

“…A large body of evidence suggests that Fz has a key role in sensing positional information; although what is upstream of Fz remains unknown. It has been proposed that ft and ds control PCP upstream of Fz/Dsh pathways, and that a gradient of Ft activity sets up the initial asymmetrical localization (Ma et al, 2003;Rawls et al, 2002;Yang et al, 2001), but the origins of the gradient of Ft activity are unknown. One of the most appealing models of positional information is based on the presence of a morphogen-like molecule that would activate Fz in a dose-dependent manner (Adler et al, 2000;Fanto et al, 2003).…”

“…We therefore isolated and characterized the expression patterns of several genes encoding members of the PCP pathway in Platynereis. The global module of the PCP pathway includes the atypical cadherins Fat and Dachsous (Ds), and their regulator Four-jointed (Fj), a Golgiassociated kinase 45,46 . This module establishes a polarity at the scale of the tissue via the opposite transcriptional gradients of ds and fj 45,46 .…”
