Fibrosis and bone marrow: understanding causation and pathobiology

Kanjaksha Ghosh,
Durjoy K. Shome,
Bipin Kulkarni
et al.

Abstract: Bone marrow fibrosis represents an important structural change in the marrow that interferes with some of its normal functions. The aetiopathogenesis of fibrosis is not well established except in its primary form. The present review consolidates current understanding of marrow fibrosis. We searched PubMed without time restriction using key words: bone marrow and fibrosis as the main stem against the terms: growth factors, cytokines and chemokines, morphology, megakaryocytes and platelets, myeloproliferative di… Show more

“…Osteoblasts originate from MSCs and contribute to bone formation. MSCs can initiate tissue fibrosis by mobilizing a subset of specialized immunophenotypes to transform into fibroblasts ( Ghosh et al, 2023 ), and also by transporting miRNAs through secretory vesicles. Wnt/β-catenin signaling has been recognized as a pathway for osteogenic differentiation ( Arya, Saranya & Selvamurugan, 2024 ).…”

“…The mechanisms underlying phenotypic variations among MPNs with a similar mutation profile require further study, as reviewed by Ghosh et al [87]. The VAF of the driver mutations may play a significant role.…”

“…Primary myelofibrosis (PMF) is a rare form of myeloproliferative tumor (prevalence 1-9:100,000) in which clonal hyperproliferation of stem cell-derived mature myeloid lineages (erythrocytes, leukocytes, megakaryocytes) are associated with bone marrow fibrosis (caused by abnormal collagen and/or reticulin production) causing a dramatic decrease in erythropoiesis and angiogenesis [1]. Furthermore, osteosclerosis, atypic megakaryocytes, extramedullary hematopoiesis, and abnormal cytokine expression are additional characteristics of PMF patients [2].…”
