Fibronectin-Containing Extracellular Vesicles Protect Melanocytes against Ultraviolet Radiation-Induced Cytotoxicity

Abstract: Skin melanocytes are activated by exposure to UV radiation to secrete melanin-containing melanosomes to protect the skin from UV-induced damage. Despite the continuous renewal of the epidermis, the turnover rate of melanocytes is very slow, and they survive for long periods. However, the mechanisms underlying the survival of melanocytes exposed to UV radiation are not known. Here, we investigated the role of melanocyte-derived extracellular vesicles in melanocyte survival. Network analysis of the melanocyte ex… Show more

“…Each of these cell types has an important function within the skin. For example, the neural crest derived melanocytes produce melanin that affords protection from UV damage (Bin et al, 2016). Langerhans cells are a type of dendritic cell immune cell that protects against infection (Berberich et al, 2003).…”

“…Regarding the impact of EVs on skin homeostasis, keratinocyte-derived EVs enhance melanin synthesis by upregulating melanosomal protein [9]. Conversely, melanocyte-derived EVs protect melanocytes and keratinocytes from UV irradiation in an autocrine or paracrine manner [8,10]. Propionibacterium acnesor Staphylococcus aureus-derived EVs are associated with acne vulgaris and atopic dermatitis, respectively [11,12].…”

“…Cells at this stage also showed upregulated expression of factors associated with advanced (IL-8, NF-κB) or very advanced (MMP1) HDF senescence (Supplementary Figure S1) [19]. We isolated EVs from 10 6 young (PDL < 10) and 10 6 senescent (PDL > 50) HDFs by conventional ultracentrifugation [8]. Senescent HDFs produced considerably more EVs (6.49 ± 0.83-fold) than young HDFs based on protein concentration ( Figure 1C).…”
