Fibroblast growth factor receptor 1 and 3 expression is associated with regulatory PI3K/AKT kinase activity, as well as invasion and prognosis, in human laryngeal cancer

Abstract: From our data we conclude that FGFR1 and FGFR3, as well as its downstream regulatory PI3K/AKT kinases, may serve as potential biomarkers for the invasiveness and prognosis of laryngeal cancer. The expression of FGFR1/3-PI3K/AKT regulatory pathway members may be instrumental for the identification of patients at risk for an unfavorable clinical outcome.

“…PI3K/AKT signaling is a well-characterized drug target for its central role in mediating cancer cell survival and proliferation. 33,34 Hyper-activation of PI3K/AKT cascade is essential for cell proliferation and survival of glioma cells. 35 Inhibition of PI3K/AKT pathway led to cell apoptosis and cell growth arrest in glioma.…”

“…FGFR‐mediated activation of RAS/MAPK/ERK is initiated by phosphorylation of FRS2α, which also leads to activation of PI3K/AKT signaling . PI3K/AKT kinase activity rather than ERK activation may play a key role in certain cancers addicted to FGFR . Rogaratinib caused inhibition of downstream phosphorylation of ERK as well as AKT with similar nM potency in cell lines overexpressing either FGFR1 (DMS‐114) or FGFR2 (MFM‐223).…”

“…FGF10 is known to activate the Erk and AKT signaling pathways (52,53). FGF22 is an important paralog of FGF10 (54,55), and FGFR1 and FGFR3 are involved in the activation of Erk and AKT signaling (56,57). TGFB3 is a secreted ligand of the transforming growth factor ␤ (TGF-␤) superfamily.…”
