Fibrinolysis and angiogenesis in wound healing

Abstract: The healing wound offers a clear example of the sequence of events in chronic inflammation leading to repair. Although angiogenesis has an obvious and essential role in this process, it has been little studied. For an angiogenic factor to seem relevant, it would have to be shown to precede the peak of increased vascularity. To define this peak, the vessel content of simple, incised mouse wounds was estimated using morphometry of histological sections, and found to rise to a maximum at days 5 and 6. Total angio… Show more

“…As these results suggested that the MEST-encoded protein may be involved in the process of neoangiogenesis that occurs during tumour formation, Mest expression was assessed during wound healing of murine skin. Although neoangiogenesis occurs during wound healing (Kjolseth et al, 1994;Thompson et al, 1991), Mest transcripts could not be detected in sections of skin undergoing healing (not shown).…”

“…It involves the activation, migration, and proliferation of endothelial cells from preexisting venules. It can be influenced by factors like hypoxia, matrix components, metabolic gradients and growth factors 1,2 . Growth factors driving re-epithelialization are central to the wound-healing process.…”

“…During the wound healing processes, an abundant blood supply is necessary to meet the enormous local demands of debridement, fibroblast proliferation, extracellular matrix synthesis, and epithelialization 1,2,3 . Impairment of blood supply may be a contributing factor in delayed healing, or nonhealing, in chronic wounds such as diabetic foot ulcers, pressure ulcers, and wounds caused by chronic and acute arterial occlusion 4,5 .…”

“…Wound healing can be considered to be composed of three overlapping events: first, inflammation; second, new tissue formation; and third, matrix remodeling [2]. In the second step, angiogenesis is essential for the provision of oxygen and metabolites to tissues [3]. We previously reported that A. vera gel stimulated angiogenesis in a chick embryo chorioallantoic membrane (CAM) assay [4] and that it increased the proliferation of calf pulmonary artery endothelial cells [5].…”
