Fibrinogen Fucosylation as a Prognostic Marker of End-Stage Renal Disease in Patients on Peritoneal Dialysis

Abstract: Glycosylation may strongly affect protein structure and functions. A high risk of cardiovascular complications seen in patients with end-stage renal disease (ESRD) is, at least partly associated with delayed clot formation, increased clot strength, and delayed cloth lysis. Taking into consideration that fibrinogen mediates these processes, we isolated fibrinogen from the plasma from patients with ESRD on peritoneal dialysis (ESRD-PD), and examined glycosylation of native fibrinogen and its subunits by lectin-b… Show more

“…Protein precipitation and affinity methods have been used for fibrinogen isolation from human plasma to investigate its N-glycosylation in association with diseases. , The former exploits the low solubility of fibrinogen (among the major plasma proteins) and depends on the dehydration of proteins, which likely results in the coprecipitation of fibrinogen with different proteins, particularly albumin . Both protein precipitation and corona enrichment are not as selective as the affinity method; however, the major advantage of the corona enrichment method for glycan profiling compared to the precipitation method is that it could be utilized in different contexts, for different corona protein enrichment by changing the surface chemistry or plasma protein/NP ratio.…”

“…DD was higher in patients with CAPD than normal volunteers [16]. PD patients have higher fibrinogen concentrations than HD patients because glucose‐based dialysate produces glycation end products [6, 17]. DD is especially higher in patients with PD [18].…”

“…Više od 90% svih smrtnih ishoda se dešava u poznim godinama (gerijatrijska populacija), a češća je kod imunokompromitovanih osoba (primarne ili sekundarne imunodeficijencije) ili kod transplantiranih bolesnika zbog primene imunosupresivne terapija, kao i kod bolesnika u stanjima hronične inflamacije, kao što su bolesnici lečeni različitim hemioterapeuticima ili različitim metodama zamene bubrežne funkcije eng.
