FGF23-regulated production of Fetuin-A (AHSG) in osteocytes

Abstract: Our data not only confirm that AHSG is produced in bone, mainly in osteocytes, but show for the first time that its production is modulated by FGF23. Since both proteins play important roles in the bone and cardiovascular pathology, these results add new pieces to the puzzling relationship between bone and vascular pathology, in particular in CKD patients, prompting future investigations in this field.

“…31 These genes encode for fibroblast growth factor 23 (FGF23), the members of short integrin-binding ligand-interacting glycoprotein (SIBLING) family of proteins DMP1 and matrix extracellular phosphoglycoprotein (MEPE), phosphate-regulating gene with homologies to endopeptidases on the X-chromosome (Phex), and fetuin-A (Figure 2 and 3). 31, 49 …”

“…There were no emPAI scores that significantly and positively correlated with age. AHSG, also known as fetuin-A, is produced not only in the liver but also in osteocytes [47], and is characterized by particularly high mineral affinity. IGFBP5 is an IGF-binding protein involved in the regulation of cell growth and is the most abundant IGFBP stored in bone, where it controls osteoblast differentiation and osteoblast–osteoclast crosstalk [48].…”

“…In a prospective cohort of 3860 patients with CKD stages 2-4, enhanced FGF-23 levels were associated with higher risk of CVD, especially with CHF [72]. FGF-23 seems to regulate the production of fetuin-A, a glycoprotein with anti-calcification activity [73]. Considering the crosstalk between these two proteins, both appear as promising renocardiovascular biomarkers.…”
