FGF Signaling in Lung Development and Disease: Human Versus Mouse

Abstract: Fibroblast growth factor 10 (FGF10) plays an important role in mouse lung development, injury, and repair. It is considered the main morphogen driving lung branching morphogenesis in rodents. While many studies have found FGF10 SNPs associated with COPD and branch variants in COPD smokers, there is no evidence of a causative role for FGF10 or these SNPs in human lung development and pediatric lung diseases. We and others have shown divergent roles for FGF10 in mouse lung development and early human lung develo… Show more

“…In this context, the abovementioned RTKs not only allow the cells to retain mesenchymal properties but also serve as markers of the adoption of an alternative phenotypic state. For instance, similar to EGFR TKI-resistant tumor cells, normal lung fibroblasts express high levels of FGFR and PDGFR, indicating the critical importance of these growth factor signals in survival of mesenchymal cells (Green et al, 2016;Danopoulos et al, 2019). Therefore, in contrast to the conventional paradigm that separates EMTassociated resistance to EGFR TKIs from bypass-dependent disease, these phenomena can be united as two sides of a phenotypic switch driven by chronic EGFR inhibition (Figure 2).…”

“…Some review articles have mentioned the involvements of TGF-β/BMP, Wnt, sonic hedgehog, and FGF10 signaling in lung development and diseases [93][94][95][96][97]. The signaling pathways of these molecules are regulated by miRNAs.…”

“…The crosstalk between the epithelial and mesenchymal compartment is conveyed by several diffusible signals from multiple signaling pathways including Sonic Hedgehog (SHH) [32], Wingless-related Integration Site (WNT) [33], Transforming Growth Factor β (TGFβ), Bone Morphogenetic Protein (BMP) [34], Fibroblast Growth Factor (FGF) [35], Hippo [36], and Retinoic Acid [4], just to name a few. In the next sections, we will dissect the role of RA throughout the five lung developmental phases, namely, embryonic, pseudoglandular, canalicular, saccular, and alveolar ( Figure 2), and also in lung vascular development.…”
