Fetal–maternal communication: the role of Notch signalling in embryo implantation

Abstract: The establishment of a successful pregnancy requires the implantation of a competent blastocyst into a 'receptive' endometrium, facilitating the formation of a functional placenta. Inadequate or inappropriate implantation and placentation is a major reason for infertility and is thought to lead to first-trimester miscarriage, placental insufficiency and other obstetric complications. Blastocystendometrial interactions are critical for implantation and placental formation. The Notch signalling family is a recep… Show more

“…It is reported that EMT is induced by NOTCH signaling, an intracellular signaling pathway induced by cell attachment (Gonzalez & Medici 2014). On blastocyst implantation, NOTCH signaling is achieved via cell-cell interaction, in which NOTCH receptors expressed on the surface of endometrium interact with ligands present on the surface of blastocysts in humans and mice (Cuman et al 2014). Activin A or TGF-β upregulates several transcription factors such as SNAI2, ZEB1 or TWIST1 mediated by phosphorylated SMAD2 or SMAD3 (Lamouille et al 2014, Li et al 2015.…”

“…Notch signaling is critical for maternal-fetal communication during implantation and placentation (9). Our laboratory has previously reported that both the conditional deletion of Notch1 in the mouse uterus and NOTCH1 silencing in Human Uterine Fibroblasts (HuFs) inhibit decidualization (10,11).…”

“…Notch1-3 are additionally expressed by the trophectoderm of human blastocysts (Aghajanova et al, 2012). Notch ligands and receptors are localized to the apical surface of the luminal epithelium during the midsecretory phase of the menstrual cycle, where they are accessible to ligands and receptors on the trophectoderm, in order to facilitate endometrial-embryo attachment (Cuman et al, 2014).…”
