Fetal liver endothelium regulates the seeding of tissue-resident macrophages

Abstract: Macrophages are required for normal embryogenesis, tissue homeostasis and immunity against microorganisms and tumours. Adult tissue-resident macrophages largely originate from long-lived, self-renewing embryonic precursors and not from haematopoietic stem-cell activity in the bone marrow. Although fate-mapping studies have uncovered a great amount of detail on the origin and kinetics of fetal macrophage development in the yolk sac and liver, the molecules that govern the tissue-specific migration of these cell… Show more

“…They occur in endothelial cells (EC) of capillaries and venules in select vascular beds (reviewed in). The diaphragms are thin protein barriers associated with endothelial specific microdomains (ie fenestrae, transendothelial channels, caveolae and vesiculo‐vacuolar organelles) with roles in the maintenance of vascular permeability, blood/tissue homeostasis and the immune function …”

“…During later embryonic stage, macrophages can be produced by fetal liver monocytes. Seeding of fetal liver monocytes in different tissues depends on plasmalemmal vesicle-associated protein [22]. Macrophages can also be derived from bone marrow-derived monocytes at steady state.…”
