Abstract:Simplifying and improving our understanding of the protocols for inducing or synchronizing estrus is important for improving management of dairy cattle. This study evaluated the estrus response and conception rate of crossbred (Holstein Friesian X Zebu) dairy cows (n=75) and heifers (n=47) assigned to a 10-day controlled internal drug release (CIDR) deviceprostaglandin F2-alpha (PGF2α) -equine chorionic gonadotrophin (eCG) based estrus synchronization protocol. Animals were assigned to three groups (a) anestrus (n=62), (b) repeat-breeders (n=11) and (c) silent estrus (n=46), according to their reproductive history, per rectal palpation, and progesterone concentrations determined by On-Farm enzyme-linked immunosorbent assays (ELISA) in milk (lactating cows) or serum (dry cows and heifers). For each animal, a CIDR device (containing 1.38g progesterone) was inserted into the vagina and allowed to remain in-situ for 10 days. On Day 8 after device insertion, a dose of 500µg of PGF2α was administered intramuscularly (IM). On Day 10 (device removal), 500 IU of eCG was given IM. Animals were inseminated or mated 48 and 72 hrs post device removal. Pregnancy was assessed 20-24 days post insemination by measurement of progesterone in milk/serum and pregnancy was confirmed by per rectal palpation and/or ultrasonography 44-90 days post insemination. Overall estrus response and conception rates were 97.5% and 78.3%, respectively, with no significant differences in parity, pre-treatment reproductive status and farming system (smallholder vs organized commercial farms). Conception rate after induced/synchronized estrus was 82.9% and 72.1% in cows and heifers, respectively, with an overall calving rate of 94.4%. In conclusion, the present study has shown high estrus response and conception rate in crossbred dairy cattle using this CIDR -PGF2α -eCG based estrus synchronization protocol. Application of this protocol is highly recommended to enhance fertility of dairy cattle in the study area, and this could readily be applied to other regions.

Fixed-time artificial insemination using injectable progesterone: ovarian follicular dynamics and pregnancy rates of Nelore cows (Bos indicus) with and without a corpus luteum

This study evaluated the follicular dynamics and the pregnancy rates of Nelore cows with or without a CL synchronized with FTAI employing injectable progesterone (P4). Multiparous cows (n=44), 45-65 days postpartum were allocated into a 2x2 factorial. Eleven cows with and 11 without a CL received a conventional protocol of FTAI (D0/1 g P4 + 2 mg EB; D8/500 μg cloprostenol, 300 IU eCG and 24 h later 1 mg EB; D10/FTAI). Other 11 cows with and 11 without a CL received 350 mg of injectable P4 (300 mg sc and 50 mg im) and 2 mg of EB im on D0. On D6, 500 μg of cloprostenol and 300 IU of eCG were administered im. After 24 h, 1 mg of EB was administered im and FTAI was performed on D8. The results were analyzed by ANOVA and Tukey or by Chi-square test (p≤0.05). The presence of follicles ≥ 5mm on D4, follicular growth rate and CL diameter were similar between groups (50-55%, 1.1-1.2mm/ day and 16.1-17.6mm; respectively). The diameter of the dominant follicle (DF) at FTAI and ovulatory follicle (OF) were similar between groups with CL (10.5±1.8 and 11.4±1.3mm, respectively) and without CL (10.8±2.3 and 11.1±2.2mm, respectively) but device was higher than injectable (11.7±2.0 and 11.8±1.7 vs. 9.7±1.7 and 10.6±2.0mm, respectively). The ovulation and pregnancy rate were similar between groups with CL (59% and 36%, respectively) and without CL (77% and 27%, respectively) but device was higher than injectable (91% and 45% vs. 45.5% and 18%; respectively). The ciclicity did not affect the follicular dynamics and pregnancy rate. The device showed higher diameter of the DF at FTAI, OF and ovulation and pregnancy rates. However, the injectable P4 produced similar percentages of follicles ≥ 5 mm on D4, follicular growth rate and CL diameter, raising the possibility of further investigation. The aim of this study was to verify the efficacy of reusing intravaginal progesterone (P4) devices on the reproductive parameters in Santa Inês ewes. Females received intravaginal P4 devices for their first, second or third use for five days plus 300 IU eCG IM and 5mg dinoprost laterovulvar 24h before device removal. Blood was collected at different moments. Transrectal ultrasonography was performed from device removal to ovulation. Part of the ewes were submitted to artificial insemination by laparoscopy (IAL -n=55) with fresh semen, whereas the rest were bred by fertile rams (n=41). On the initial 18 h, ewes that received devices for the first time showed higher P4 concentrations (5.1±1.8 vs 3.5±1.4 vs 2.4±1.1 -P<0.05). However, after the first 48h no difference was observed among all treatments and P4 supraluteal concentrations were detected in all ewes upon device removal. Estrous response, interval from device removal to estrus, rate of ovulating animals, number of ovulations, time from device removal to ovulation and average conception rates after IAL or natural mating were similar among all 3 groups.
