Ferrous iron oxidation and rusticyanin in halotolerant, acidophilic ‘Thiobacillus prosperus’

Abstract: The halotolerant acidophile ‘Thiobacillus prosperus’ was shown to require chloride for growth. With ferrous iron as substrate, growth occurred at a rate similar to that of the well-studied acidophile Acidithiobacillus ferrooxidans. Previously, the salt (NaCl) requirement of ‘T. prosperus’ was not clear and its growth on ferrous iron was described as poor. A subtractive hybridization of cDNAs from ferrous-iron-grown and sulfur-grown ‘T. prosperus’ strain V6 led to identification of a cluster of genes similar to… Show more

“…prosperus V6 (DSM 14174) that has similarity to the rus operon of At. ferrooxidans T (Nicolle et al, 2009) and it is hypothesized that expression of the rusticyanin gene is actively involved in Fe oxidation, presumably in a similar way to that described for At. ferrooxidans T .…”

“…ferrooxidans T is inhibited by low concentrations of chloride (Blake et al, 1991; Harahuc et al, 2000), whereas chloride is required for expression of rusticyanin in Ac. prosperus V6 (Nicolle et al, 2009) and maximum iron oxidation in Ac. prosperus T was seen at 20 g/L NaCl.…”

“…('Thiobacillus prosperus') still awaits resolution. The main distinguishing feature of this acidophile is its tolerance to salt, being able to grow in up to 3.5 % (0.6 M) sodium chloride, whereas most iron-oxidizing acidithiobacilli are inhibited by 1 % salt (Nicolle et al, 2009). Both the original strain and a novel strain (V6) have been shown to grow optimally in the presence of 1-2 % sodium chloride and also, like Af.…”
