Ferroptosis: a new regulatory mechanism in neuropathic pain

Lu Li,
Lingling Guo,
Rui Gao
et al.

Abstract: Neuropathic pain (NP) is pain caused by damage to the somatosensory system. It is a common progressive neurodegenerative disease that usually presents with clinical features such as spontaneous pain, touch-evoked pain, nociceptive hyperalgesia, and sensory abnormalities. Due to the complexity of the mechanism, NP often persists. In addition to the traditionally recognized mechanisms of peripheral nerve damage and central sensitization, excessive iron accumulation, oxidative stress, neuronal inflammation, and l… Show more

“…Peripheral nerve injury induces oxidative damage caused by excessive ROS production in the spinal cord, leading to disruption of cell-membrane structure [ 61 ]. Activation of microglia, in addition to the release of inflammatory factors, leads to the overproduction of ROS in the spinal cord [ 62 , 63 ], thus disrupting neuronal biofilms and inducing disturbances in glycerophospholipid metabolism.…”

“…Of note also is autophagy, although the related mechanisms remain indefinable, and further investigation is necessary [9]. Interesting also is the contribution of ferroptosis, a novel cell death form caused by iron-dependent lipid peroxidation [10][11][12] and associated with the pathogenesis of many diseases, such as ND. Such has led some to suppose that variations in the iron metabolism's homeostasis, the consequent induction of oxidative stress, and the related inflammation in the CNS is involved in the onset of ferroptosis and neuronal health [10][11][12].…”

In such a process, the apelinergic system, mediated by ELA/APJ signaling, has also been recently documented to participate in this regulation [13].…”
