Female Sex Hormones and Cardiac Pressure Overload Independently Contribute to the Cardiogenic Dementia Profile in Yucatan Miniature Swine

Abstract: Post-menopausal women with heart failure (HF) frequently exhibit cardiogenic dementia. Using a pre-clinical swine model of post-menopausal HF, we recently demonstrated that experimental menopause (ovariectomy; OVX) and HF (6-month cardiac pressure overload/aortic banding; AB) independently altered cerebral vasomotor control and together impaired cognitive function. The purpose of this study was to examine the prefrontal cortex and hippocampus tissues from these animals to assess whether OVX and HF are associat… Show more

“…Additionally, ERK activation has been reported in OVX studies. 90,91 Given DHA's proposed influence upon both PI3K-AKT-mTOR 92 and ERK signaling pathways, 93 the altered DHA/PUFA status associated with VCD treatment, and the subsequent reduction in BDNF may offer a plausible mechanistic basis for their disruption, although further confirmatory experiments are required. As discussed mTOR and ERK are central components of synaptic plasticity-related signaling and are involved in various down-stream pathways explaining the diminished transcription of the CREB family transcription factors (Creb1 and Atf4), both of which have integral roles in synaptic plasticity.…”

“…Animal models provide more direct evidence of the effects of a Western diet on brain insulin signalling and AD markers. Miniature swine fed a high-fat, high-fructose corn syrup, highcholesterol diet for 10 weeks demonstrated reduced insulinstimulated Akt signalling in the prefrontal cortex (Hayward et al 2019). Further, hamsters and wildtype mice display reduced hippocampal and cortex insulin signalling when fed either an SFA and sucrose diet (Mielke et al 2006) or sweetened water (60% fructose or sucrose) (Mielke et al 2005).…”

“…We used our published protocol to isolate soluble Aβ peptides 13 , 74 . Briefly, samples (20–30 mg wet weight) were homogenized in 20 volumes of ice-cold RIPA buffer containing a protease inhibitor cocktail and centrifuged at 12,000× g (10 min; 4 °C).…”
