Female Mice Expressing Constitutively Active Mutants of FSH Receptor Present with a Phenotype of Premature Follicle Depletion and Estrogen Excess

Abstract: Strong gain-of-function mutations have not been identified in humans in the FSH receptor (FSHR), whereas such mutations are common among many other G protein-coupled receptors. In order to predict consequences of such mutations on humans, we first identified constitutively activated mutants of the mouse (m) Fshr and then expressed them under the human anti-Müllerian hormone promoter in transgenic mice or created knock-in mutation into the mouse genome. We show here that mutations of Asp580 in the mFSHR signifi… Show more

“…Nardo and other scholars [20][21][22][23][24] examined the expression of integrin ανβ3, α4β1 in the endometrium of the fertile women 6 day and 8 days after ovulation, ανβ3, α4β1 was found to be transiently expressed during the implantation window. Meanwhile, the expression of integrin ανβ3 in pregnant women in the implantation window was much higher than that in their non-pregnant counterparts.…”

“…In addition to determination of thyroid status, other hormones affecting bone can be assayed using commercial assays, including corticosterone, vitamin D, parathyroid hormone, and insulin-like growth factor-1. In addition, sex hormone and gonadotropin status can be determined if required (735). Indirect analysis of bone turnover can be obtained by measurement of bone resorption and formation markers using commercially available kits (725).…”
