Feedback Control of Gene Expression Variability in the Caenorhabditis elegans Wnt Pathway

Abstract: Variability in gene expression contributes to phenotypic heterogeneity even in isogenic populations. Here, we used the stereotyped, Wnt signaling-dependent development of the Caenorhabditis elegans Q neuroblast to probe endogenous mechanisms that control gene expression variability. We found that the key Hox gene that orients Q neuroblast migration exhibits increased gene expression variability in mutants in which Wnt pathway activity has been perturbed. Distinct features of the gene expression distributions p… Show more

“…MYC output would be expected to fluctuate with the loss of positive and negative feedback from FBP, FIR, and FUSE. 56 The increased variation in Myc expression elicited by FBP KO mirrors the variation in MYC levels seen in XP. 6 In XPB disease, the responses of the 3 0 to 5 0 p89/XPB helicase of TFIIH to effector domains of FBP and FIR are mutationally disabled, and the output of MYC is buffeted by the changes in the flux of the pathways that converge onto the MYC locus at the level of the chromatin and transcription machineries.…”

“…Theoretical and experimental efforts have provided strong evidence that variability in gene expression plays a significant role in controlling cellular phenotypes in development (49), health (50) and disease (51,52). While the transcriptional mechanisms responsible for controlling this variability continue to be an active area of research (53), the potential system-level interactions are less well explored, and network analysis of GRNs is a powerful approach to fill this gap.…”

“…For example, the mab-5 gene in C. elegans is a known target of Wnt signaling [93], but a W-CRM in its regulatory DNA had not been identified [94]. Using our search protocol, we identified a FF7 pair 9.4 kB upstream of the mab-5 ATG, which was demonstrated by others to have W-CRM activity in mab-5 expressing cells [94]. Expression of this reporter was significantly reduced by mutation of the HMG site identified by our search [94].…”
