FDA-Approved Amoxapine Effectively Promotes Macrophage Control of Mycobacteria by Inducing Autophagy

Abstract: The emergence and spread of multidrug-resistant (MDR) and extensive drug-resistant (XDR) TB urges the development of new therapeutics. One promising approach to combat drug resistance is targeting host factors necessary for the bacteria to survive or replicate while simultaneously minimizing the dosage of traditional agents.

“…90 Various drugs, including isoniazid and pyrazinamide (first-line tuberculosis drugs) and other compounds and plants mentioned in previous studies, have been reported to target and activate host autophagy. 91,92 Recent studies have shown a variety of old remedies to be remarkably effective in warding off infections, including clonamine, 91 the antidepressant drug amoxapine, 93 and so on. Berbamine (BBM), an FDA-approved drug, inhibits drug-sensitive and resistant Mtb growth by promoting macrophage autophagy.…”

“…In contrast, the antiprotozoal drug nitazoxanide has been found to strongly stimulate autophagy while inhibiting mTOR signaling 377 . Additionally, the antidepressant amoxapine has demonstrated the ability to induce autophagy and protect macrophages during infection 378 . The anticonvulsant drugs carbamazepine and valproic acid, meanwhile, can induce mTOR‐independent autophagy through AMPK activation, a unique characteristic that sets them apart from other drugs 379 .…”
