Fatty liver induced in Zucker "fatty" (ff) rats by a semisynthetic diet rich in sucrose.

Abstract: A new fatty liver model is described in which heredity plays a major role. A marked fatty liver develops when the homoz gous mutant Zucker rat (ff or "fatty") is fe a semisynthetic Jiet enriched in sucrose. Lean littermates do not develop fatty livers on this experimental diet. Even old ff rats do not develop fatty livers on chow; the experimental diet is required. The fatty liver is described, including light microscopic and preliminary electron microscopic observations. The clinical importance of understandi… Show more

“…( 14 ) The constituents of this diet induce gradual onset of hepatic steatosis compared with previously reported diets. ( 14 ) Rats were divided into three experimental groups: (i) rats fed the diet for 12 weeks (D12 group, n = 7), (ii) rats fed the diet for 12 weeks and treated with LPS (LPS group, n = 7), (iii) rats fed the diet for 24 weeks (D24 group, n = 7). Rats in the LPS group were injected intraperitoneally (i.p.)…”

“…14 Fischer 344 rats also develop steatosis on a high-sucrose/fructose diet. Zucker fatty (fa/fa) rats are particularly sensitive to fatty livers induced by feeding 60% sucrose 62 or orotic acid (1%). In all strains of rats that have been evaluated, males are more susceptible to fatty liver than females.…”

“…Intracellular assembly of VLDL in hepatocytes appears to involve two separate steps: 1 ) cotranslational binding of apoB to a small quantity of triglycerides to form a nascent lipid-poor complex in the rough endoplasmic reticulum, and 2 ) transport of the lipid-poor particles to the smooth endoplasmic reticulum, where more triglycerides are added to form the larger, triglyceride-rich VLDL particles (45)(46)(47)(48)(49). The assembly and secretion of VLDL in the liver can be regulated by several factors, including the availability of intracellular lipids (especially triglycerides) (50,51), expression levels of microsomal triglyceride transfer protein (52)(53)(54)(55), and dietary and hormonal effects (56)(57)(58).…”
