Fatty Acid Modulated Human Serum Albumin Binding of the β-Carboline Alkaloids Norharmane and Harmane

Abstract: Harmane and norharmane are representative members of the large group of natural β-carboline alkaloids featured with diverse pharmacological activities. In blood, these agents are transported by human serum albumin (HSA) which has a profound impact on the pharmacokinetic and pharmacodynamic properties of many therapeutic drugs and xenobiotics. By combination of various spectroscopic methods, the present contribution is aimed to elucidate how nonesterified fatty acids (FAs), the primary endogenous ligands of HSA… Show more

“…It forms a π-π stacking interaction with Y411 at 3.52 Å ( Fig 5C ). This binding is similar to the binding behaviour of nonharmane [ 35 ] and Bcl2 family anticancer agent [ 36 ] as previously reported. In addition to this, the FA4 backbone stabilization role of π-alkyl interactions with Subdomain IIIA residues V426 at 4.91Å and L460 at 5.17 Å is observed.…”

“…Free BC can enter the brain, whereas the albumin bound fraction cannot. Albumin binding of BC requires, however, a unique co-binding of free fatty acids 10 . French fries have been found to correlate inversely with ALS risk 7 , whereas a very low body mass index, in particular in adolescence and young adulthood is carrying a risk to develop the disease later in life 1,2 .…”

“…Interaction of various NMs with the most abundant human serum albumin (HSA) has been investigated [ 74 , 75 ]. Systematic studies on the interaction of the main drug binding components of human plasma HSA or alpha 1 -acid glycoprotein (AGP) with NMs may possibly influence not only the free concentrations of exogenous and endogenous ligands [ 76 , 77 ], however, the biodistribution of NMs as well.…”
