Fatty Acid Composition of Serum Lipid Fractions in Type 2 Diabetic Patients With Microalbuminuria

Abstract: OBJECTIVE -To determine the fatty acid composition of serum phospholipid, triglyceride, and cholesterol ester fractions and to analyze the lipid profile of microalbuminuric type 2 diabetic patients. RESEARCH DESIGN AND METHODS-A case-control study was conducted with 72 patients: 37 were normoalbuminuric (urinary albumin excretion rate [UAER] Ͻ20 g/min), and 35 were microalbuminuric (UAER 20 -200 g/min). After 4 weeks of a standardized diet, the fatty acid composition of phospholipid, triglyceride, and choleste… Show more

“…DNA was isolated from lymphocytes using standard procedures (16). Subjects were genotyped for FABP2 A54T polymorphism as previously described (12). All PCRs were run in a final volume of 25 l, containing 50 ng of genomic DNA, 20 mmol/l Tris-Cl, (pH 8.4), 50 mmol/l KCl, 1.5 mmol/l MgCl 2 , 0.2 mmol/l dNTPs, 1 unit of TaqDNA polymerase, and 1 mol/l of specific primer.…”

“…Serum saturated fatty acids might induce endothelial dysfunction (10) and are related to increased cardiovascular mortality (11). We have previously reported increased levels of serum saturated fatty acids in patients with type 2 diabetes and microalbuminuria (12), as well as a reduction in albumin excretion rate after replacement of red meat (high content of saturated fatty acids) with chicken (low content of saturated fatty acids) in these patients (13). Based on these observations, we hypothesized that genetically predisposed subjects exposed to a diet with a high content of saturated fatty acids might be at high risk for developing renal disease.…”

“…In our experiments, 15 metabolites involved in different biochemical metabolic pathways, such as phospholipids, FFAs, and steroid hormones, were distinguishable between the healthy control and 2 patient groups. Earlier studies have demonstrated that diabetes is intimately associated with metabolic disorders of lipids, such as phospholipids (21,22 ). Phospholipids are important constituents of biomembranes that participate in various biological pathways (23,24 ) and are therefore a particular focus of attention in metabolomic studies.…”

“…Serum FA profi le associates with IL-1 ␤ expression in AT In the analysis of serum and AT FA composition in different lipid fractions, we concentrated on the TG fraction due to the fact that the FA profi le in this fraction refl ects both the diet and endogenous FA metabolism ( 20 ). Furthermore, the strongest correlations between serum FA composition and tissue IL-1 ␤ expression were observed in the TG fraction.…”
