Oil seeds are a natural source of phytochemicals with high nutritional value. The present study aims to evaluate for the first time the antioxidant, enzyme inhibitory, and anti-inflammatory activities in vitro and in vivo, as well as the physicochemical properties, fatty acid compositions, total polyphenol, flavonoid, and tocopherol contents of Algerian Prunus amygdalus var. amara cold-pressed oil. Bitter almond produces seeds that yield 35.5% of oil. It was characterized by a high saponification index of 215.94 mg KOH g −1 of oil, a peroxide value of 7.74 meq O 2 kg of oil, a K 232 of 2.612, and a K 270 of 0.39. This oil represented a valuable source of healthy fatty acids. GC-MS analysis revealed oleic (68.27%) and linoleic (16.14%) as the main fatty acids. α-Tocopherol (85.77 mg/kg) was found to be the major component. The total phenolic and flavonoid contents were 21.94 ± 0.29 and 21.52 ± 0.14 µg/mL, respectively. The oil showed good antioxidant activity (A 0.50 = 34 ± 0.44 μg/ mL) using the CUPRAC assay and modest activity with DPPH, ABTS, FRAP, and β-carotene assays, respectively. BAO (bitter almond oil) displayed the most promising α-glucosidase and α-amylase activities compared to acarbose as a reference molecule and moderate acetylcholinesterase (AChE) inhibitory activity. Moreover, BAO demonstrated in vitro and in vivo anti-inflammatory efficacy in a dose-dependent manner, comparable to that of diclofenac sodium. These effects were confirmed by histological examinations. Overall, our results showed that BAO has a strong potential to design new industrial preparations with nutritional, pharmaceutical, and cosmeceutical applications.

The aim of this study is to determine the phenolic composition of Hypericum linarioides Bosse, to assess its various biological properties, and finally to confirm the experimental data of enzyme inhibitions via docking studies. LC-MS/MS analysis performed over methanol extract (ME) revealed the presence of 27 phytochemicals. Antioxidant activity results indicated that ME had the strongest DPPH radical scavenging and ferric ion reducing capacity than water and hexane extracts. XTT assays revealed the strongest anti-proliferative activity of ME against MCF-7 and HT-29 cancer cells. Flow cytometry results also confirmed the notable apoptotic effect of ME on MCF-7 cells through mitochondria-dependent pathway. Enzyme activity results exhibited the most promising acetylcholinesterase and α-glycosidase inhibitory activities of the ME. Molecular docking calculations of α-glycosidase with the major compounds (1, 15, 17 and 19) indicated that 17 (isoquercitrin) had the best docking binding energy (À 10.92 kcal/mol) with appropriate binding interactions.

Five 4‐hydroxyphenylethyl substituted pyridine enhanced, precatalyst, preparation, stabilization, and initiation‐Pd‐N‐heterocyclic carbene (PEPPSI‐Pd‐NHC) complexes are synthesized in a straightforward way. All PEPPSI‐Pd‐NHC complexes were prepared by mixing 4‐hydroxyphenylethyl substituted NHC precursors, palladium chloride, potassium carbonate, and potassium bromide in pyridine. All complexes were screened for human carbonic anhydrase I (hCA I) and hCA II, acetylcholinesterase (AChE), butyrylcholinesterase (BChE), and α‐glucosidase (α‐Glu) inhibitory activities. The ChE inhibitory activities of the new PEPPSI‐Pd‐NHC complexes bearing the 4‐hydroxyphenylethyl group (1a–e) against α‐Glu, AChE, and BChE were determined by the Tao and Ellman methods. The results indicated that all the synthetic complexes exhibited potent inhibitory activities against all targets as compared to the standard inhibitors, revealed by IC50 values. The Ki values of the new PEPPSI‐Pd‐NHC complexes 1a–e for hCA I, hCA II, AChE, BChE, and α‐Glu were obtained in the ranges of 18.98–32.65, 22.95–38.13, 3.67–11.65, 4.09–9.36, 186.92–287.45 µM, respectively. Among the synthesized complexes, the most potent complexes were 1c toward hCA I and II with Ki values 18.98 and 22.95 µM, and 1d toward AChE and BChE with Ki = 3.67 and 4.09 µM, respectively.
