Fatty acid‐binding protein 5 controls lung tumor metastasis by regulating the maturation of natural killer cells in the lung

Abstract: Fatty acid-binding protein (FABP) 5 is highly expressed in various types of tumors and is strongly correlated with tumor growth, development, and metastasis. However, it is unclear how the expression of FABP5 in the host affects tumor progression. In this study, using a lung tumor metastasis model in mice, we found that FABP5-deficient mice were more susceptible to tumor metastasis, which is accompanied by infiltration of a lower frequency of activated natural killer (NK) cells in the lung. Additionally, FABP5… Show more

“…The scRNA-seq results show that only FABP3/4/5/7, and a very small amount of FABP6, are expressed in NSCLC. In this malignancy, FABP5 was reported to facilitate tumorigenesis and metastasis [ 52 , 53 ], while the functions of FABP3 and 4 are less clear and even become controversial [ 54 ], and the role of FABP6 and 7 remain unclarified in NSCLC. In fact, our work firstly reports the pro-metastatic function of FABP7 in NSCLC.…”

“…Studies has provided evidences that PSAT1 is elevated in NSCLC. PSAT1 overexpression impairs the prognosis of cancer by promoting cancer cell proliferation, metastasis, as well as chemoresistance discovered that NRF2 regulates the expression of the important serine synthesis enzymes PHGDH, PSAT1, and SHMT2 through the activation of ATF4 to promote glutathione and nucleotide formation [ 114 ]. In response to serine deprivation, ATF4 stimulates serine biosynthesis genes in NSCLC cells [ 115 ].…”

“…Reported mechanisms underlying these effects are quite varied and include protein-protein interaction and modulation of HIF-1α ( 153 ) or estrogen-related receptor (ERR)-α ( 152 ), regulation of TNF-α-dependent NF-κB signaling ( 180 ) or PI3 kinase/Akt signaling ( 114 ), and regulation of lipid metabolism ( 18 , 52 ) or of angiogenesis via VEGFA-dependent signaling ( 138 ). It is noteworthy, however, that several reports indicate that inhibition/knockdown of FABP5 could have an opposite effect, resulting in more skin tumors ( 199 ) and increased lung cancer metastases ( 194 ).…”
