Fasting Launches CRTC to Facilitate Long-Term Memory Formation in Drosophila

Abstract: Canonical aversive long-term memory (LTM) formation in Drosophila requires multiple spaced trainings, whereas appetitive LTM can be formed after a single training. Appetitive LTM requires fasting prior to training, which increases motivation for food intake. However, we found that fasting facilitated LTM formation in general; aversive LTM formation also occurred after single-cycle training when mild fasting was applied before training. Both fasting-dependent LTM (fLTM) and spaced training-dependent LTM (spLTM)… Show more

“…It has been reported that appetitive long-term memory (LTM) is consolidated more quickly than aversive LTM [36,37]. Moreover, Hirano et al showed that different LTM systems are utilized in the hungry state [38]. The rats used for MAZE testing in this study were kept at a mildly hungry state, thus different effects from GEN exposure may have occurred.…”

“…Many molecules that are specifically required for LTM have been identified, and some of these molecules are involved in learning-dependent gene transcription/ translation (3,(53)(54)(55)(56)(57). For instance, fasting-dependent LTM that is formed without training repetition requires CREB-regulated transcription coactivator (CRTC)-mediated cAMP response element binding protein (CREB) transcription in the MB (12). Such a transcription-dependent mechanism takes time and may thus underlie gradual formation of memory induced by the ltm-PAM.…”

“…Recent studies have revealed aspects of the molecular and cellular mechanisms of LTM formation induced by repetitive training (8)(9)(10)(11), but how a single training induces a stable LTM is poorly understood (12).…”

“…The exposure of flies to repetitive training cycles with gaps between the training cycles (spaced training) leads to a consolidated form of long-term memory that is CREB transcription dependent and lasts up to a week. Training without gaps (massed training) leads to the formation of anesthesia-resistant memory (ARM), which similar to long-term memory, is typically measured 24 hr after 5 cycles of training 7,13,[15][16][17]20,21 .…”

“…The length of the adult stage accommodates longer-term genetic, behavioral, dietary and pharmacological manipulations of memory, in addition to determining the effect of aging and neurodegenerative disease on memory [3][4][5][6][11][12][13][15][16][17][18][19][20][21] . Classical conditioning is induced by the simultaneous presentation of a neutral odor cue (conditioned stimulus, CS + ) and a reinforcement stimulus, e.g., an electric shock or sucrose, (unconditioned stimulus, US), that become associated with one another by the animal 1,16 .…”
