Fast, ultra-trace detection of juvenile hormone III from mosquitoes using mass spectrometry

Abstract: In the present work, a new protocol for fast separation and quantification of JH III from biological samples using liquid chromatography coupled to electrospray tandem mass spectrometry is described. In particular, the proposed protocol improves existing methodologies by combining a limited number of sample preparation steps with fast LC-MS/MS detection, providing lower limits of detection and demonstrated matrix effect control, together with high inter and intraday reproducibility. A limit of detection of 8 p… Show more

“…A 10 μl aliquot of deuterated JHIII analog (6.25 ppb) in acetonitrile was added to each sample before extraction with 600 μl hexane. The organic phase was dried under a stream of nitrogen and resuspended in 50 μl of acetonitrile and subjected to analysis by HPLC-MS/MS analyses [39].…”

“…We have previously described the potential of mass spectrometry (MS) based protocols for structural identification and quantification of JH homologues from insect samples 11,12 . More recently, we reported on a liquid chromatography-mass spectrometry (LC-MS/MS) protocol for the identification and quantification of the five most common JH homologs 13 .…”

“…Hemolymph was obtained by perfusion 13 , and JH III titers were measured using a high performance liquid chromatography coupled to electrospray tandem mass spectrometry approach (HPLC-MS/MS) 14 . The changes in the titers of JH detected in hemolymph of sugar-fed females after adult emergence, as well as following a blood meal are presented in Fig.…”
