Fast-response fluorescent probe with favorable water solubility for highly sensitive imaging of endogenous tyrosinase in living cells and zebrafish model

Photoacid sulfonato-merocyanine (MEH) serving as a photoisomer is added in mixed solution of amphiphilic calix[4]arene (SC4A8) and tetra-(4-pyridylphenyl) ethylene (TPE-4Py), which not only activates the in situ supramolecular self-assembly or disassembly, but also regulates two kinds of fluorescence emission. Benefiting from that MEH releases protons and becomes a ring-closed isomer spiropyran (SP) under 420 nm irradiation, TPE-4Py is protonated and then assembled with SC4A8 to form a supramolecular assembly with yellow fluorescence at 540 nm via electrostatic interactions. In the dark, SP changes to ring-opened MEH, realizing a spontaneous disassembly accompanied by fluorescence emission at 495 nm, and thus, a photo-activated reversible in situ supramolecular system is obtained successfully. Furthermore, Rhodamine B (RhB) acting as an energy acceptor is doped in the ternary supramolecular assembly, enabling the efficient energy transfer by the light irradiation. The formed tunable multicolor supramolecular assemblies have been successfully applied to the information encryption by 3D color codes.

Aluminum (Al) is widely used in food industry, but excessive absorption of Al3+ causes many serious health problems. Therefore, it is of great significance to determine the specificity of Al3+. In this study, we designed and synthesized a water‐soluble perylene diimide derivative (PDI‐Phe) for monitoring Al3+ rapidly and visually. Through absorption and emission spectroscopy techniques, we systematically explored the properties and mechanism of the sensor to Al3+. The limit of detection (LOD) of Al3+ was as low as 6.54 nM. And with the existence of Al3+, electrostatic interaction and intermolecular π‐π stacking promoted the aggregation and fluorescence quenching of PDI‐Phe. Even in the presence of other analytes, PDI‐Phe still had excellent selectivity and sensitivity for Al3+ detection. Besides, it also had the characteristics of low cost, good water solubility, excellent chemical and optical stability, and easy synthesis. Finally, this method can be further applied to tap water samples.

The design of dual‐mode fluorescence and Raman tags stimulates a growing interest in biomedical imaging and sensing applications as they offer the possibility to synergistically combine the versatility and velocity of fluorescence imaging with the specificity of Raman spectroscopy. Although lanthanide‐doped fluoride nanoparticles (NPs) are among the most studied fluorescent nanoprobes, their use for the development of bimodal fluorescent‐Raman probes has never been reported yet, to the best of the authors knowledge, probably due to the difficulty to functionalize them with Raman reporter groups. This gap is filled herein by proposing a fast and straightforward approach based on aryl diazonium salt chemistry to functionalize Eu3+ or Tb3+ doped CaF2 and LaF3 NPs by Raman scatters. The resulting surface‐enhanced Raman spectroscopy (SERS)‐encoded lanthanide‐doped fluoride NPs retain their fluorescence labeling capacity and display efficient SERS activity for cell bioimaging. The potential of this new generation of bimodal nanoprobes is assessed through cell viability assays and intracellular fluorescence and Raman imaging, opening up unprecedented opportunities for biomedical applications.
