BackgroundJujube extract is commonly used as a food additive and flavoring. The sensory properties of the extract, especially sweetness, are a critical factor determining the product quality and therefore affecting consumer acceptability. Small molecular carbohydrates make major contribution to the sweetness of the jujube extract, and their types and contents in the extract have direct influence on quality of the product. So, an appropriate qualitative and quantitative method for determination of the carbohydrates is vitally important for quality control of the product.ResultsHigh performance liquid chromatography-evaporative light scattering detection (HPLC-ELSD), liquid chromatography-electronic spay ionization tandem mass spectrometry (LC-ESI-MS/MS), and gas chromatography–mass spectrometry (GC–MS) methods have been developed and applied to determining small molecular carbohydrates in jujube extract, respectively. Eight sugars and alditols were identified from the extract, including rhamnose, xylitol, arabitol, fructose, glucose, inositol, sucrose, and maltose. Comparisons were carried out to investigate the performance of the methods. Although the methods have been found to perform satisfactorily, only three sugars (fructose, glucose and inositol) could be detected by all these methods. Meanwhile, a similar quantitative result for the three sugars can be obtained by the methods.ConclusionsEight sugars and alditols in the jujube extract were determined by HPLC-ELSD, LC-ESI-MS/MS and GC–MS, respectively. The LC-ELSD method and the LC-ESI-MS/MS method with good precision and accuracy were suitable for quantitative analysis of carbohydrates in jujube extract; although the performance of the GC–MS method for quantitative analysis was inferior to the other methods, it has a wider scope in qualitative analysis. In situ analysis of odor is an important approach to connect odor with chemical composition. However, it is difficult to conduct a rapid direct analysis of the odor sample because of low analyte concentration and sampling. To achieve the direct analysis, a carbon fiber ionization mass spectrometry (CFI-MS) method has been developed and applied to measure volatile components releasing from intact jujube. To build the CFI source, a 2.0-cm long carbon fiber bundle was integrated on the pin of a commercial corona discharge needle by mean of a 1.3-cm long stainless hollow tube. Odor sample driven by N2 gas can be directly introduced to the carbon fiber bundle to complete the ionization of analytes. Acetic acid, ethyl acetate, ethyl caproate, octyl acetate, and damascone present in jujube were selected to evaluate the performance of the CFI-MS method on quantitative analysis of the gaseous sample. Good lineary was obtained (R2 ≥ 0.9946) between 5.0 and 500.0 ng/L with limits of detection (LOD) ranging from 0.5 to 1.5 ng/L. Recoveries of five volatile compounds for the spiked jujube samples were between 94.36 and 106.74% with relative standard deviations (RSDs) less than 7.27% (n = 5). Jujube of different varieties can be distinguished by principal components analysis based on the analytical results of volatile compounds. The developed method demonstrated obvious advantages such as simplicity, high throughput, good sensitivity and wide range of applicability, which will be an alternative way for in situ analysis of the odor sample.

A simple method was established for the rapid determination of low molecular carbonyl compounds by the combination of atmospheric pressure chemical ionization tandem mass spectrometry (APCI-MS/MS) and data mining. The ionization was carried out in positive mode, and six low molecular carbonyl compounds of acrolein, acetone, propionaldehyde, crotonaldehyde, butanone, and butyraldehyde were analyzed by both full scan mode and daughter scan mode. To overcome the quantitative difficulties from isomer of acetone/propionaldehyde and butanone/butyraldehyde, the quantitation procedure was performed with the characteristic ion of [CH3O]+ under CID energy of 5 and 15 eV. Subsequently, the established method was successfully applied to analysis of six low molecular carbonyl compounds in tobacco smoke with analytical period less than four minutes. The contents of acrolein, acetone, propionaldehyde, crotonaldehyde, butanone, and butyraldehyde for a cigarette were about 63 ± 5.8, 325 ± 82, 55 ± 9.7, 11 ± 1.4, 67 ± 5.9, and 12 ± 1.8 μg/cig, respectively. The experimental results indicated that the established method had the potential application in rapid determination of low molecular carbonyl compounds.
