Fas-Mediated Inflammatory Response inListeria monocytogenesInfection

Abstract: The molecular mechanisms of Fas (CD95/Apo-1)-mediated apoptosis are increasingly understood. However, the role of Fas-mediated production of proinflammatory cytokines such as IL-18 and IL-1β in bacterial infection is unclear. We demonstrate the importance of Fas-mediated signaling in IL-18/IL-1β production postinfection with Listeria monocytogenes without the contribution of caspase-1 inflammasome. IL-18/IL-1β production in L. monocytogenes–infected peritoneal exudate cells from Fas-deficient mice was lower th… Show more

“…Interestingly, recent studies have found that caspase-8 is involved in modulating IL-1b maturation (42)(43)(44)(45)(46)(47). Caspase-8 is also involved in priming inflammasome for activation during Salmonella infection (31).…”

“…However, RIPK3 has been shown to be partially involved in this TRIF–caspase 8-dependent pathway 108 . Engagement of FAS ligand (FASL; also known as TNFSF6 and CD95) by FAS is another pathway by which caspase 8 is activated to cleave IL-1β and IL-18 independently of caspase 1 (Refs 109, 110 ). In this context, TLR-induced FAS expression drives caspase 8- and FADD-mediated release of IL-1β and IL-18 (Ref 109 ).…”

“…However, the FasL-induced release of IL-18 and IL-1β does not require caspase-1, NLRP3, or NLRC4 [42,43]. Very recently, caspase-8, but not caspase-1, caspase-9, or caspase-11, was demonstrated to be critical for Fas-mediated IL-18 maturation [44,45]. As mentioned above, IL-18 can induce FasL expression by NK cells [37].…”
