Familial risk of migraine: A population‐based study

Abstract: Estimates of familial aggregation of migraine have varied considerably due, in part, to methodological differences among studies. We concluded a population-based study of 73 clinically confirmed probands with migraine, 72 matched control probands, and 511 of their first-degree relatives, all of whom were directly interviewed. The risk of migraine was 50% more likely in relatives of migraine probands than in relatives of controls. Migraine risk was considerably higher among relatives of probands with disabling … Show more

“…Risk factors for having the condition of migraine-An increased risk of having migraine headache has been noted among people who have a first degree relative with migraine [43,48,50] and between twins, with a pair-wise concordance rate being significantly higher in monozygotic twins (43%) compared to dizygotic twins (12%) [51]. This suggests that there is some underlying genetic factor(s) that predispose people to developing the condition of migraine headaches.…”

“…6 Migraine frequently runs in families, but family and segregation studies have produced conflicting results with respect to the mode of inheritance. [7][8][9][10] From a large proband-oriented clinical study 6 it was inferred that migraine with aura is largely or exclusively determined by genetic factors, whereas migraine without aura seems to be caused by a combination of both genetic and environmental factors. Thus migraine is a multifactorial disorder.…”

“…15 From a pathophysiological perspective, central neuronal hyperexcitability underlies susceptibility to and the development of migraine episodes. 16,17 The broad evidence base for this as an aetiology suggests a multifactorial causation, with amino acids, calcium channels, magnesium depletion and controlling genes all being implicated.…”
