FADS Gene Polymorphisms, Fatty Acid Desaturase Activities, and HDL-C in Type 2 Diabetes

Abstract: Polyunsaturated fatty acids (PUFA) correlate with risk of dyslipidemia and cardiovascular diseases. Fatty acid desaturase (FADS) single nucleotide polymorphisms (SNPs) modulate circulating PUFA concentrations. This study examined influence of FADS1 and FADS2 genetic variants on desaturase activities and blood lipid concentrations in type 2 diabetes patients, and further assessed their interrelationships. Selected SNPs (FADS1: rs174547, rs174548, rs174550; FADS2: rs174575, rs174576, rs174583, rs498793 and rs272… Show more

“…Polymorphisms in these genes influence the circulating levels of various metabolic forms of n-3 and n-6 fatty acids including eicosanoid precursors (217). Multiple studies have found that carriers of the C allele in the FADS1 gene (rs174547) had reduced endogenous production of LC-PUFAs (51,52,218). SNPs in the FADS2 gene (rs2727270, rs498793) have also been correlated with altered PUFA metabolism (51,52).…”

“…Each additional copy of the rs9939609 A allele has been associated with a BMI increase of a mean of 0.10 Z -score units, equivalent to ~0.4 kg/m 2 (Sonestedt et al, 2009 ; Tanofsky-Kraff et al, 2009 ; Zhao et al, 2014b ). For the observed genotypes in APOA2 and FADS1 , there is no associated effect (Yabuta et al, 2016 ; Huang et al, 2017 ; Ching et al, 2019 ; Moran et al, 2019 ; Amengual et al, 2020 ; Graßmann et al, 2020 ; Wang et al, 2020 ). For LEPR , a study found that rs1137101 AG and GG carriers with a high fat total intake had 3.0 times higher risk of obesity and 4.1 times higher risk of high cholesterol levels than those with a low intake of total fat (Domínguez-Reyes et al, 2015 ).…”

“…Fads2 (involved in biosynthesis of polyunsaturated fatty acids (Huang et al, 2017;Reynolds et al, 2018)), were upregulated and downregulated, respectively, in a temporal manner. In addition, different signalling arms were modulated distinctly in IF16 and EOD.…”
