Factors underlying the variability of lipid droplet fluorescence in MA‐10 leydig tumor cells

Abstract: Neutral lipids accumulate in cellular lipid droplets. These droplets vary remarkably in number and amount between cells. In the present studies, the variability in lipid content was quantified by comparing the coefficient of variation of fluorescence histograms of nile red lipid-stained cells to the variability of cell size or cell protein distributions. This measure of lipid droplet variability persisted through a wide range of cell lipid content and averaged 4.4-fold more variability than cell size and 2.6-f… Show more

“…This was investigated by analyzing the fluorescence intensity and distribution of the hydrophobic probes Bodipy and Nile red in macrophages. 31,32 When apoptotic cells were incubated with Bodipy, a lipidinteracting fluorescent molecule, the accumulation of the probe in the cytosol increased as deduced by confocal microscopy ( Figure 4A). This was confirmed by flow cytometry of cells treated for 18 h with LPS and IFNg, or for 8 h with GSNO.…”

“…Vesicles containing neutral lipids were stained with the fluorescent dye BODIPY 493/503 (Molecular Probes number D3922) as described (32,33). This BODIPY fluorophor is a lipophilic dye that has an affinity for neutral lipids and is not a fatty acid analog.…”
