Resumo -O objetivo deste trabalho foi padronizar e caracterizar as condições para determinação da atividade da redutase do nitrato em tecido foliar de cana-de-açúcar, com uso do método in vivo. Amostras foliares foram coletadas de uma lavoura de primeira soqueira da cultivar IACSP 933046, com idade de seis meses. Foram estudadas diferentes condições de preparo das amostras foliares e do meio de incubação. O material que possibilitou a maior atividade da redutase do nitrato foi obtido pela amostragem de 25 discos de 1 cm de diâmetro, coletados às 13h, do centro da folha do tipo +1 sem nervura. O meio de incubação otimizado para a determinação da atividade dessa enzima em folhas de cana-de-açúcar deve ser composto por: 2,5 mL de KNO 3 300 mmol L -1 ; 2,5 mL de tampão fosfato 285 mmol L -1 pH 7,3; 1,0 mL de Tween 20 a 0,6% (v/v); e 4,0 mL de água deionizada. A maior atividade da redutase do nitrato é obtida pela incubação das amostras por 90 min, a 32°C, no escuro; é observada em plantas jovens formadas pela brotação da soqueira; e alcança o valor mínimo na fase de maturação das plantas.Termos para indexação: Saccharum, atividade enzimática, estádio fenológico, método in vivo, nitrogênio, ritmo diurno. Optimization of nitrate reductase activity analysis and its characterization in sugarcane leavesAbstract -The objective of this work was to standardize and characterize the conditions for determining the nitrate reductase activity in sugarcane leaf tissue, using in vivo method. Leaf samples were collected from a first ratoon crop of the IACSP 933046 cultivar, with six months of age. Different preparation conditions of leaf samples and incubation media were studied. The material that allowed the highest nitrate reductase activity was obtained by the sampling of 25 discs of 1 cm in diameter, collected at 13h from the center of the +1 leaf type without center rib. The incubation medium optimized to determine nitrate reductase activity in sugarcane leaves must be comprised of: 2.5 mL KNO 3 300 mmol L -1 ; 2.5 mL phosphate buffer 285 mmol L -1 pH 7.3; 1.0 mL Tween 20 0.6% (v/v); and 4.0 mL deionized water. Cytokinin (CK) is a vital plant hormone that controls many aspects of growth and development in plants. Nitrogen (N) is the indispensable macronutrient needed in plants and also one of the most important limiting factors for plant growth. This study was designed to investigate the simultaneous effects of CK and N on the visual turf quality and antioxidant metabolism of drought-stressed creeping bentgrass (Agrostis stolonifera L.). ‘PennA-4’ creeping bentgrass treated with trans-zeatin riboside at three rates of CK concentrations of 0, 10 and 100 μM (designated by CK0, 10, and 100) and two nitrogen rates with 2.5 and 7.5 kg N·ha-1 every 15 days (designated by low and high N) in a complete factorial arrangement was grown under two soil moisture regimes: well-watered and drought stress. Exogenous CK improved turf quality and delayed leaf wilting under drought stress, especially under high N. The grasses treated with CK10 and CK100 had lower O2- production and H2O2 concentration than those without CK treatment. The CK100 treatment enhanced the activities of superoxide dismutase (SOD), ascorbate peroxidase (APX), catalase (CAT), and guaiacol peroxidase (POD) by 25%, 22%, 17% and 24%, respectively, relative to CK0. Moreover, the activity changes of the antioxidant enzyme isoforms were more significant under high N condition relative to low N condition. Our results demonstrated the beneficial impacts of CK and N on physiological reactions, especially antioxidant metabolism, and foliar application of CK at 10 or 100 μM plus 7.5 kg ha-1 N biweekly may improve drought stress resistance of creeping bentgrass.

Creeping bentgrass {Agrostis stolonifera L.) is a major low-cut turfgrass species worldwide. Information is limited on the diurnal variation in its N metabolism, particularly the impacts of cytokinin (CK) and nitrate (NO3"). To gain insight into the diurnal fluctuation of N metabolism and the effects of CK and nitrate, the transcript abundance of the major N metabolism enzymes in shoots, including nitrate reductase (NR), nitrite reductase, chloroplastic glutamine synthetase (GS2), ferredoxin-dependent glutamate synthase (Fd-GOGAT), and glutamate dehydrogenase, were studied during the day/ night cycle (14/10 h). All measured parameters exhibited clear diurnal changes with some differences among treatments (control, CK at 50 MM, and nitrate at 7.5 kg N ha-'), except GS2 expression and total protein. Both NR expression and nitrate content showed a peak at 0430 h (8.5 h in dark). Nitrate nutrition increased diurnal change of nitrate content, showing an over two-fold diurnal difference compared to 63 and 81% differences for the control and CK treatment, respectively. Simultaneously, CK shifted the diurnal in vivo NR activity pattern. In addition, creeping bentgrass appears to be able to maintain N assimilation at night comparable to that during the day, demonstrated by either similar or even higher levels of measured parameters between certain phases of light and dark periods. This study provides further information for understanding how CK and nitrate regulate N metabolism in creeping bentgrass.
