Facilitating Akt Clearance via Manipulation of Hsp70 Activity and Levels

Abstract: Members of the 70-kDa heat shock family can control and manipulate a host of oncogenic client proteins. This role of Hsp70 in both the folding and degradation of these client proteins makes it a potential drug target for certain forms of cancer. The phenothiazine family of compounds, as well as the flavonoid myricetin, was recently shown to inhibit Hsp70-ATPase activity, whereas members of the dihydropyrimidine family stimulated ATPase function. Akt, a major survival kinase, was found to be under the regulatio… Show more

“…These effects of PPZ were linked by biochemical, cell biological, and genetic studies to activation of PP2A, a tumor-suppressive protein phosphatase. The ability of PPZ to activate PP2A thus represents the likely mechanism for the previously unexplained ability of phenothiazines to downregulate PI3K/AKT signaling and may explain the recurrent identification of these compounds in unbiased screens for agents with antineoplastic activity (18)(19)(20)(21)(49)(50)(51)(52)(53).…”

“…The antipsychotic effects of phenothiazines correlate with their ability to block dopamine receptors, but a broad array of other activities have been described, including antitumor effects. The basis for the antiproliferative activities of phenothiazines are uncertain and have been variously attributed to a number of mechanisms, including inhibition of PKC (17), calmodulin (18), PI3K/AKT signaling (18)(19)(20)(21), and cancer stem cell activity (22).…”

“…Because Hsc70 largely functions as a protein "foldase" (35), and Tau folding is thought to be an early event in disease progression (11,12,36), we investigated what impact Hsc70 could be having on Tau folding.…”

“…HSP90 and HSP27 are among the binding partners of Akt with similar effects where HSP27 promotes the binding of activating kinases and HSP90 prevents dephosphorylation. HSP70 on the other hand causes Akt degradation (Koren et al 2010;Konishi et al 1997;S. Sato, Fujita, and Tsuruo 2000).…”

“…It has very diverse functions within the cell (Mayer 2013). It aids the folding of proteins, prevents aggregation, involved in quality control, disassembly of protein complexes, stabilizing regulatory proteins, cell cycle regulation, aiding signal transduction and apoptosis (Kirkegaard et al 2010;Koren et al 2010;J. C. Young 2010).…”
