Facilitated endospore detection for Bacillus spp. through automated algorithm‐based image processing

Abstract: Bacillus spp. endospores are important dormant cell forms and are distributed widely in environmental samples. While these endospores can have important industrial value (e.g. use in animal feed as probiotics), they can also be pathogenic for humans and animals, emphasizing the need for effective endospore detection. Standard spore detection by colony forming units (CFU) is time-consuming, elaborate and prone to error. Manual spore detection by spore count in cell counting chambers via phase-contrast microscop… Show more

“…These shells are made of a proteinaceous coat, consisting of S-layer proteins, aminopeptidases and flagellin [23]. This coating allows the bacteria to survive harsh external conditions to germinate in the gut, making them suitable for use in animal feed as probiotics [24]. Once sporulated in the gastrointestinal tract, these probiotics can release a wide range of active substances including enzymes, nutrients and antimicrobial compounds [25].…”

“…Their idea was to use ML in order to have a more efficient sample throughput with high accuracy [57]. Biermann et al used an algorithm-based image processing for the at-line determination of endospores from microscopic pictures [79]. Here, too, the aim was to achieve a faster result using ML.…”

“…After an eight step process the autolysis of the mother cell releases the matured endospore [1,2,[6][7][8][9]. This cell type is largely dehydrated [10] and includes a typical biomarker called dipicolinic acid [6,[11][12][13]. Especially the nutrient depletion acts as the main trigger for sporulation and, thus prevents active growth and sporulation at the same time [1,3].…”
