Facile Deposition of Manganese Dioxide to Albumin-Bound Paclitaxel Nanoparticles for Modulation of Hypoxic Tumor Microenvironment To Improve Chemoradiation Therapy

Abstract: Tumor microenvironment with hypoxia and excess hydrogen peroxide (HO) tremendously limits the effect of chemoradiation therapy of colorectal cancer. For the first time, we developed a facile method to deposit manganese dioxide (MnO) on the surface of albumin bound paclitaxel nanoparticles (ANPs-PTX) to obtain MnO-functioned ANPs-PTX (MANPs-PTX). In the tumor microenvironment, MANPs-PTX could consume excess hydrogen peroxide (HO) to produce abundant oxygen for tumor oxygenation and improve chemoradiation therap… Show more

“…In this study, for the purpose of delivering extremely hydrophobic PFTBA in vivo, albumin was picked out as the ideal drug carrier. It is well known that albumin-based nanoparticles have been widely used in the clinic, like Abraxane (28). Of critical importance, Fluosol-43 (a PFTBA nanoemulsion) with superior biosafety has been already approved by the US Food and Drug Administration as an oxygen carrier to improve myocardial oxygenation (29,30).…”

“…In addition, due to its good chemical oxygen-carrying capacity, hemoglobin is also widely used to relieve tumor hypoxia, but it can cause acute nephrotoxicity and acute hypertension, which pose a serious threat to patient lives 10 , 11 . Currently, tumor environment-responsive oxygen delivery strategies, such as manganese dioxide, have also been shown to be effective in reversing hypoxic tumors to a certain extent 5 , 12 . However, the tumor re-oxygenation levels achieved by this method are still seriously restricted by the limited amount of H 2 O 2 in the tumor surroundings and the toxicity to normal cells of Mn 2+ itself 13 .…”

“…It is noteworthy that Mn 2+ released from MnO 2 into the TME is an efficient MRI contrast agent. 22,23 MRI was applied for diagnosis of tumors with a 1.5 T clinical MR scanner at room temperature. Images of the three groups showed different degrees of whitening effects with the increase of the MnO 2 concentration (Fig.…”
