Ezetimibe-Statin Combination Therapy: Efficacy and Safety as Compared With Statin Monotherapy

Abstract: SUMMARYIntroduction: To date, most clinical comparisons of ezetimibe-statin combination therapy versus statin monotherapy have relied entirely on surrogate variables. In this systematic review, we study the efficacy and safety of ezetimibe-statin combination therapy in comparison to statin monotherapy in terms of the prevention of cardiovascular events in hyperlipidemic patients with atherosclerosis and/or diabetes mellitus.

“…The recommended ezetimibe dosage is 10 mg daily, with or without food, taken either 2 hours before or 4 hours after bile acid sequestrants (BAS) if used in combination. 11 , 12 When administered as monotherapy or coadministered with statins, timing of administration has no effect on their potency. This was confirmed by the study of Hyung Sik Yoon, who tested the therapeutic efficacy and safety of a fixed-dose combination of ezetimibe 10 mg and simvastatin 20 mg, in terms of morning versus evening administration.…”

“…Therefore, combination-drugs has become a promising direction to improve clinical efficacy. For instance, ezetimibe-statin combination is a more efficacy and safe therapy in clinical treating ( Nußbaumer et al, 2016 ). Unfortunately, numerous adverse effects are still a matter of concern ( Ferri and Corsini, 2020 ).…”

“…Interestingly, this pathway produces cholesterol and other isoprenoids, such as coenzyme Q 10 (CoQ) [22]. In addition to statins, other therapeutic strategies have been developed, for instance using inhibitors of the N-terminal Niemann-Pick C1-like protein 1 (NPC1L1) receptor, which is located on the luminal surface of enterocytes and is responsible for cholesterol uptake [23]. Recently, innovative therapies using not only chemical drugs, but also monoclonal antibodies against certain proteins, such as the proprotein convertase subtilisin/kexin type 9 (PCSK9) have been utilized with promising results.…”
