Classification of micronuclei induced by lomefloxacin, a difluorinated quinolone bactericidal agent, in mouse bone marrow was performed by fluorescence in situ hybridization using DNA probes for the centromere repeated minor satellite DNA and the telomeric hexamer repeat (5'-TTAGGG-3'). Colchicine and mitomycin C were used as a positive control aneugen and clastogen, respectively, and these compounds produced the expected responses. Single doses of 40, 80, 160, or 320 mg/kg lomefloxacin were given via oral intubations and bone marrow was sampled at 24 and 48 hr after treatment. The micronuclei showed significant increases in both sampling times after doses of 320 mg/kg. A statistically significant increase of micronuclei frequency was also detected for 160 mg/kg lomefloxacin at 48 hr after treatment. The responses were directly correlated with bone-marrow cytotoxicity. Following treatment with 160 and 320 mg/kg lomefloxacin, 48.2 and 50.0% of the induced micronuclei, respectively, showed double labeling with centromeric signals and several telomeric signals, indicating that the induced micronuclei were composed of whole chromosomes. Similarly, 51.8 and 50.0% of the induced micronuclei, respectively, were centromere-negative, demonstrating that lomefloxacin not only induces chromosome loss but also chromosome breakage. The results also showed that chromosomes can be enclosed in a micronucleus before and after centromere separation. Overall, this study provides the first evidence of the potential of lomefloxacin to induce aneugenic effect in mice. However, given the high doses used in this study, the clinical significance of this finding is uncertain.

In the present work the induction of micronuclei in erythrocytes of Cyprinus carpio treated with X-rays and colchicine is studied for the evaluation of mutagenic effects of both clastogenic and mitoclastic (spindle poisoning) agents in this system.Three different experiments were performed treating groups of laboratory-reproduced animals with (1) single doses of X-rays (0.1, 0.5 and 2 Gy); (2) a single i.p. injection of colchicine at the concentrations: 1.6 × 10 −2 , 8× 10 −2 , 0.4 and 2 mg/kg b.w. so as to mimic an acute exposure to the agent and (3) six repeated i.p. injections of the first three concentrations of colchicine, over a period of 18 days, so as to mimic a chronic exposure. Repeated blood samplings were performed by cardiac puncture over a period of about 2 months after the treatment and micronucleus frequencies were determined at multiple times on the same individuals after mutagen exposure. A dose-dependent increase in the micronucleus frequency was observed in irradiated fish and a peak value detected at 21 days. Slight increases of micronucleus frequencies were also observed in both colchicine experiments only for the highest concentrations at the earliest sampling time. Higher concentrations of colchicine clearly showed a lethal effect. According to the present data the micronucleus frequency induced by the highest colchicine dose is comparable to that observed after 0.1 Gy of X-ray irradiation.

Micronucleus is an abnormal structure in eukaryotic cells. It is the result of various physical and chemical factors, such as radiation and chemical drugs acting on dividing cells. It is generally believed that micronuclei are originated in backward chromosomes and fragments that have lost centromere and are formed at the end of mitosis. However, other experiments suggest that interphase cells can also form micronuclei. In this study, human peripheral blood lymphocytes were cultured in vitro and cyclophosphamide 20 ug/ml was injected at specifi c times of the cell cycle to observe the relationship between micronuclei formation and cell cycle. The experimental results confi rmed that the cell cycle micronucleus formation. KEYWORDS: cyclophosphamide; lymphocyte; cell cycle; micronucleus PrefaceIn this study, human peripheral blood lymphocyte micronucleus (MN) was induced by γ-ray, and cell cycle control, BrdU immunofl uorescence staining, cytokine B (CB) block cytoplasmic splitting were used to study the micronuclei formation of human lymphocytes and the quantitative relationship between the cell cycle. Results: There was no signifi cant change in micronucleus rate (MNF) and micronucleus cell rate (MNCF) between G0 and G1 lymphocyte irradiation and control group (P> 0.05). Compared with control group, MNCF increased signifi cantly (P <0.01). (3) Compared with the control group, MNF and MNCF increased 90 times and 70 times higher than those of the control group. CONCLUSION: MN can form MN in S and G2 phases in human lymphocyte cycle, but M phase is the main stage of MN formation.
