Measuring arsenic in urine provides important information for clinical and epidemiological studies. While many researchers have studied ways to improve sample storage and understand why arsenic species undergo species interconversion, none have investigated inline dilution as a solution for arsenic speciation sample stability. A fast inline dilution method for AsB, DMA, MMA, As III, and As V was demonstrated in this work. Inline dilution calibrations from a single stock standard were shown to have good linearity and resulted in LODs in the single digit ppt range. Inline dilutions of 30X, 50X, and 100X resulted in 0.1 s to 1.1 s variation in retention time. Manual sample preparation resulted in poor recovery (61%) for As III over a 24 h time period, which was a direct result of As III converting to As V. Inline dilution of urine spiked with As III resulted in good recovery (101%) and reduced the species interconversion of As III to As V to $1%. Accuracy to NIST SRM 2669 (arsenic in frozen urine) was found to be within reported values for the five arsenic species tested for in this method.

The importance of measuring arsenic (As) species has been appreciated for a long time mainly because of the wide spread knowledge of arsenic's toxicity and its use as a poison. Increasingly health, environmental and food regulations have been written around As species rather than total concentrations. Knowledge of As speciation is important as the chemical form of As controls its bioavailability, toxicity, mobility and therapeutic benefits. Arsenic is present as inorganic (arsenate, arsenite, thioarsenates), complexed (arsenic glutathionines and phytochelatins), low molecular weight (monomethylarsonate, dimethylarsenite, arsenobetaine, arsenocholine etc.) and high molecular weight ( arsenic hydrocarbons and arsenic phospholipids) species. In this review we cover the intergrity of As species during collection, storage, sample preparation and measurement by HPLC-ICPMS and HPLC-HG-AFS. The major conclusion is that it is essential to ensure that As species, especially in waters and sediments, are not artefacts of the preservation or extraction procedure. Most biota and sediment samples can be stored frozen (-20 o C), but the stability of water and sediment samples is matrix dependent and depends on preservation technique applied. Arsenic cannot be extracted from samples using a single set of conditions but must be optimised for each sample type. Methanol-water mixtures with microwave heating are commonly used to extract polar As species from tissues while As-lipids required a non-polar solvent. Dilute acid can be used to increase the efficiencies of extraction of hard to extract tissue As species. Freeze drying is suitable for the drying of biotic material while sediments should not be dried before analysis. Extraction efficiencies are critically dependent on particle size. Polar As species have a wide variety of ionic characteristics thus complimentary chromatographic 4 Understanding the action of a chemical species in a food stuff or nutritional supplement: As mentioned above, some food regulations require specification of the active ingredient of foodstuffs or supplements. However, another important reason for determining the chemical species is to understand the beneficial action of an element. For example, many food stuffs are promoted for their anti-carcinogenic activity [8]. Evaluation of the role of As in preventing cancers has required the active species to be identified [8,9].Increasingly health, environmental and food regulations have been written around a knowledge of As species rather than total concentrations. Arsenic Speciation and Cadmium Determination in Tobacco Leaves, Ash and Smoke

The concentrations of arsenic (As) and cadmium (Cd) in the tobacco leaves, ash and smoke of 10 kinds of cigarettes collected from different countries worldwide were determined by ICP-MS after microwave-assisted digestion. Total As and Cd concentrations in the tobacco leaves ranged from 0.20 to 0.63 and 1.8 to 9.9 mg kg -1 , respectively. By the speciation analysis of As in tobacco leaves and ash by HPLC-ICP-MS following acid extraction, arsenite [As(III)] and arsenate [As(V)] were determined and trace amounts of monomethylarsonic acid (MMAA), dimethylarsinic acid (DMAA), trimethylarsine oxide (TMAO), tetramethylarsonium (TeMA) and some unidentified As species were also found. Arsenic speciation for smoke absorbed in an aqueous solution was carried out. The sum of the As species in tobacco leaves, ash and smoke was in good agreement with the result of total As determination in each sample, and the recoveries of speciation were 100 ± 10%. The distributions and the behaviors of As species were clarified.
