Extraction of Annatto Seed Pigment by Supercritical Carbon Dioxide

Abstract: Operating pressures ranged 3000-7000 psi and temperatures 40-55°C. Maximum solubility of pure bixin was 0.003 and of annatto seed pigment 0.26 mg/g CO*. Analyses of annatto seed pigment from supercritical carbon dioxide extractions showed only cy-and P-bixin present. Only bixin was extracted from mixed samples consisting of bixin and norbixin. No thermal degradation product was detected in samples extracted within the temperature range we used. Increased temperature resulted in increased pigment/g CO, at all p… Show more

“…These results revealed that the yields of both compounds increased as the CO 2 pressure increased at a constant temperature of 40°C. This tendency is consistent with that reported for extracting bixin from annatto seeds (Degnan et al, 1991), carotenoids from sweetpotato roots (Spanos et al, 1993) and lycopene from tomato skin (Inakuma et al, 1998). This may be because an increase in SC-CO 2 density with an increasing pressure leads to increased solubility of compounds in SC-CO 2 as described in previous reports.…”

“…This result may be due to the higher polarity of chlorogenic acid than that of ␤-carotene and ␣-tocopherol because SC-CO 2 is thought to behave similarly to hexane. It was also reported that bixin was extracted with SC-CO 2 , but norbixin, which is more polar than bixin, was not extracted under the same conditions (Degnan et al, 1991). Figure 1 shows the relationship between the color value of a* and the ␤-carotene content of the powder samples treated with and without SC-CO 2 (n=9).…”

Commercial extract of oil soluble annatto pigments are obtained from the seeds by several processes such as suspension in oil, mechanical processes and solvent extraction (Preston andRickard, 1980 andDegnan et al, 1991). The commercial preparations of annatto colours with organic solvents have inconvenient of small concentration of pigments and residual toxic solvent in the product (Mckeown andMark, 1962 andPreston andRickard, 1980).…”
