Extraction and purification of ε-poly-l-lysine from fermentation broth using an ethanol/ammonium sulfate aqueous two-phase system combined with ultrafiltration

Abstract: Natural antimicrobial agent ε-poly-l-lysine could be alternative prepared by ethanol/ammonium sulfate ATPS combined with ultrafiltration from the microbial feedstock.

“…An aqueous biphasic system can also be combined with ultra-filtration for large scale ε-PL purification. In this system, ε-PL from culture broth was subjected to triplicate extraction by ethanol/ammonium aqueous two-phase system, followed by desalting by ultra-filtration, which together provided new avenues for ε-PL purification ( Chen et al, 2020 ). Although it resulted in a high purity (92.39%) ε-PL product at an acceptable recovery ratio (87.72%), this extraction method may cause some the environmental and economic problems.…”

“…Under the optimal conditions for adsorption and desorption on Amberlite IRC-50 resin, a final ε-PL with a purity of 92.4% was obtained with a 75% recovery [38]. Furthermore, an alcohol/salt aqueous two-phase system (ATPS) was also developed, in which an ATPS, consisting of 20% (w/w) ethanol and 20% (w/w) ammonium sulfate at pH 9.5, was applied to purify ε-PL from fermentation medium with triplicate extractions, and a final ε-PL product with 92.4% purity and 87.7% recovery was obtained with the combination of desalting by ultrafiltration [39].…”

“…Aqueous two-phase systems (ATPS) have been widely used in separations of biomolecules such as proteins, polypeptides, enzymes [1][2][3][4][5][6], bioactive compounds [7,8], and natural dyes [9][10][11][12][13][14][15], among others. Since both phases are mainly made up of water, they provide mild favorable conditions needed to preserve activities of biomolecules [16].…”
