Extraction and Characterization of Collagen from Sand Sea Cucumber (Holothuria scabra)

Abstract: Sand sea cucumber (Holothuria scabra) is an aquatic product that belongs to Echinodermata, a habitant in almost all Indonesian seas. The main component of the sea cucumber is protein, one of which is collagen. This study aimed to extract and characterize collagen from the species using the acid-base extraction method. The characterization of sea cucumber collagen includes molecular weight, amino acid components, Fourier transform infrared spectrophotometry, and scanning electron microscopy analysis. This study… Show more

“…The amount of type-1 collagen content in sea cucumbers and Tilapia-skin can vary depending on the type. 15 In contrast to previous studies, compared with other collagen sources such as catfish-skin, snakehead fish-skin, Tilapia-skin and from other mammalian skins, the percentage of collagen yield from sea cucumbers is slightly high. 16 The source of collagen was found to be higher in some types of sea cucumbers.…”

“…Furthermore, the ratio of the amide III wavenumber to 1,454 cm -1 were 1.13 (with EDTA) and 1.17 (without EDTA), suggesting that the collagen triple helix structure is intact [39]. EDTA can chelate mineral ions without cleaving and breaking the triple helix structure of Yellowstripe skin collagen [40]. Therefore the FTIR spectra of products have no significant difference with each other [41].…”

“…Prior to extraction, three sea cucumbers H. fuscocinerea and S. variegatus were sampled from the waters of the Thousand Islands National Park, cleaned using clean water, then the guts of the sea cucumbers were removed and cut into cubes. After being cut according to shape and size, samples of sea cucumbers were dried in an oven at 60 °C for 30 minutes (Syahputra et al, 2021). 20 g of dry sea cucumber samples were extracted by maceration method using 96% ethanol with a volume of 1:5 (w/v) for 3 times 24 hours.…”
